摘  要:以线粒体DNA D-loop部分基因序列为分子标记,研究辽河群体(LH)、大洋河群体(DYH)、鸭绿江群体(YLJ)、太湖群体(TH)4个地理群体的刀鲚(Coilia nasus)遗传多样性。结果显示:辽河群体、大洋河群体和鸭绿江群体刀鲚的单倍型多样性指数分别为0.85700、0.84400和0.85700,明显高于太湖刀鲚群体的单倍型多样性指数0.41700(P<0.05);核苷酸多样性指数分别为0.00263、0.00318和0.00351,明显高于太湖群体刀鲚的核苷酸多样性指数0.00091(P<0.05)。以上结果表明,辽宁地区刀鲚的遗传多样性较太湖群体丰富,其中鸭绿江群体遗传多样性最丰富,从侧面证明辽宁沿海地区适合刀鲚种群生长。此外,辽河、大洋河和太湖刀鲚群体的Tajima′s D值分别为-0.72673、-1.10277和-1.36240,鸭绿江刀鲚群体的Tajima′s D值为-1.76179,显著偏离中性(P<0.05),表明其种群史上可能经历过扩张。4个群体间的遗传距离为0.00177~0.00323,明显低于刀鲚与七丝鲚(Coilia grayi)、刀鲚与凤鲚(Coilia mystus)种间的遗传距离,Fst值为0.00543<0.05000,说明4个群体间未发生分化。以NJ和MP两方法构建的分子系统发育树结果支持4个群体刀鲚间未发生分化这一结果。The genetic diversity of Coilia nasus in four geographic groups(Liaohe group LH,Dayanghe group DYH,Yalujiang group YLJ,and Taihu group TH)was studied using partial mitochondrial DNA D-loop gene sequences as molecular markers.The results showed that the haplotype diversity index of Coilia nasus in Liaohe group,Dayanghe group and Yalujiang group were 0.85700,0.84400 and 0.85700,respectively,which were significantly higher than that of coilia nasus in Taihu group at 0.41700.The nucleotide diversity index were 0.00263,0.00318 and 0.00351,significantly higher than the Taihu group′s 0.00091.The above results showed that the genetic diversity of the Coilia nasus in Liaoning provincewas richer than that in Taihu group.The Yalu River population had the richest genetic diversity.These proved that the coastal areas of Liaoning were suitable for the growth of Coilia coilia population.In addition,the Tajima′s D value of Coilia nasus in Liaohe group,Dayanghe group and Taihu group was-0.72673,-1.10277 and-1.36240,respectively.The Tajima's D value(-1.76179)of Coilia nasus in Yalujiang group was significantly deviated from neutral(P<0.05),suggesting that the group may have experienced expansion in history.The genetic distance between the four groups was 0.00177~0.00323,which was significantly lower than the genetic distance between Coilia grayi and Coilia nasus,Coilia nasus and Coilia mystus,with a Fst value of 0.00543<0.05000,indicating that there was no differentiation between the four groups.The molecular phylogenetic tree constructed by the NJ and MP methods supported the result that four groups of Coilia nasus did not differentiate.
