摘 要:目的探究芦荟多糖对自身免疫性甲状腺炎(autoimmune thyroiditis,AT)小鼠辅助型T细胞亚群的影响。方法将50只健康无特定病原体的BALB/C雌性小鼠。随机分为芦荟多糖低、中、高剂量组、AT组、对照组,每组各10只。其中对照组小鼠均为健康小鼠,AT组及芦荟多糖低、中、高剂量组均为AT小鼠(使用猪甲状腺球蛋白免疫联合高碘水法建立小鼠AT模型),且芦荟多糖低、中、高剂量组采取药物治疗。比较5组小鼠的甲状腺功能指标[促甲状腺激素(thyroid‑stimulating hormone,TSH)、游离三碘甲状腺原氨酸(free triiodothyronine,FT3)、游离四碘甲状腺原氨酸(free tetraiodothyronine,FT4)]。分别比较小鼠外周血和脾脏组织的CD3^(+)CD8^(+)IL‑4^(+)(白细胞介素‑4,interleukin‑4)辅助T淋巴细胞和CD3^(+)CD8^(+)IFN‑γ^(+)(干扰素,interferon)辅助T淋巴细胞水平。结果在芦荟多糖治疗4周后,芦荟多糖低、中、高剂量组及AT组小鼠,TSH水平较对照组高,而FT3、FT4水平较对照组低(P<0.05)。治疗4周后,与芦荟多糖低、中、高剂量组比较,AT组TSH水平降低,而FT3、FT4水平升高(P<0.05)。治疗4周后,与对照组比较,AT组、芦荟多糖低、中、高剂量组小鼠外周血及脾脏组织的CD3^(+)CD8^(+)IL‑4^(+)辅助T淋巴细胞和CD3^(+)CD8^(+)IFN‑γ^(+)辅助T淋巴细胞水平升高(P<0.05)。治疗4周后,与AT组比较,芦荟多糖低、中、高剂量组小鼠外周血及脾脏组织的CD3^(+)CD8^(+)IL‑4^(+)辅助T淋巴细胞和CD3^(+)CD8^(+)IFN‑γ^(+)辅助T淋巴细胞水平降低(P<0.05)。结论芦荟多糖可有效改善AT小鼠的甲状腺功能,同时还可降低辅助型T细胞亚群的表达。Objective To explore the effects of aloe vera polysaccharide on helper T cell subsets in autoimmune thyroiditis(AT)mice.Methods Fifty healthy female BALB/C mice without specific pathogens were selected as research subjects,and were randomly divided into the low,medium and high dose aloe vera polysaccharide groups,each consisting of 10.The thyroid function indexes,such as thyroid‑stimulating hormone(TSH),free triiodothyronine(FT3)and free tetraiodothyronine(FT4)]were compared between the five groups.The levels of CD3^(+)CD8^(+)IL‑4^(+)(interleukin‑4),helper T lymphocytes and CD3^(+)CD8^(+)IFN in peripheral blood and spleen of the mice were also compared.Results After 4 weeks of treatment with aloe vera polysaccharide,following results were obtained:(1)The TSH levels in the mice of the low,medium and high dose aloe vera polysaccharide groups and the AT group were higher than that in the control group,while the levels of FT3 and FT4 were lower than those in the control group(P<0.05).(2)The TSH level in the AT group decreased considerably as compared with the low,medium and high dose aloe vera polysaccharide groups,while the levels of FT3 and FT4 significantly increased(P<0.05).(3)The levels of CD3^(+)CD8^(+)IL‑4^(+)helper T lymphocytes and CD3^(+)CD8^(+)IFN in the peripheral blood and spleen tissues of the mice in the AT group and the low,medium and high dose aloe vera polysaccharide groups increased as compared with those of the control group(P<0.05).(4)The levels of CD3^(+)CD8^(+)IL‑4^(+)helper T lymphocytes and CD3^(+)CD8^(+)IFN in the peripheral blood and spleen tissues of the mice in the low,medium and high dose aloe vera polysaccharide groups increased to a less extent,as compared with those of the AT group(P<0.05).Conclusion Aloe vera polysaccharide could effectively improve the thyroid function of the autoimmune thyroiditis mice,and at the same time could also reduce the expression level of helper T cell subsets,with good results.
