摘 要:目的:研究深绿卷柏总黄酮(SLJB⁃HT)的提取及其对痴呆小鼠行为的影响。方法:通过溶剂回流、大孔树脂柱色谱等现代提取分离技术,制备了深绿卷柏总黄酮部位,经改良的紫外分光光度法测定总黄酮的含量。构建D⁃半乳糖(D⁃gal)和氯化铝(AlCl_(3))诱导痴呆小鼠模型,将8周龄昆明雄性小鼠分为对照组(control)、痴呆模型组(D⁃gal,120 mg/kg,i.p+AlCl_(3),10 mg/kg,i.g,D⁃gal+Al)和处理组(SLJB⁃HT,80 mg/kg,i.g+D⁃gal+AlCl_(3))。模型组和处理组小鼠腹腔注射D⁃半乳糖(120 mg/kg),AlCl_(3)(10 mg/kg)灌胃,正常对照组腹腔注射生理盐水,双蒸水灌胃,每天一次。从第4周开始处理组小鼠每天SLJB⁃HT混悬液(80 mg/kg)灌胃,其余两组双蒸水灌胃,每天一次,连续4周。末次给药后利用旷场实验检测动物焦虑样行为,新物体识别和Y迷宫实验检测其识别记忆以及空间记忆能力。结果:(1)深绿卷柏总黄酮中的黄酮含量为43.3%(433 mg/g)。(2)D⁃gal和AlCl_(3)联合给药(D⁃gal+Al)可以诱导痴呆小鼠模型。统计分析结果显示:与对照组相比,模型组小鼠在旷场中间区的活动减少,而处理组(SLJB⁃HT)小鼠在旷场中间区的活动则明显多于痴呆小鼠,且处理组小鼠在旷场中间区移动距离的百分比明显高于模型组小鼠(P<0.05),表明处理组小鼠的焦虑程度较低。(3)与对照组和处理组相比,模型组小鼠与旧物体的互动更多。小鼠在新奇臂中的时间百分比(%),模型组的新异臂探索时间小于对照组(P<0.05),但与处理组相比差异不显著。对照组与处理组的交替正确率均高于模型组。与模型组相比,处理组的新物体识别指数明显升高,探索行为正确率也明显增加(P<0.01)。结论:SLJB⁃HT可以减轻痴呆小鼠模型脑的记忆和认知缺陷,并降低焦虑水平。Objective:To investigate the extraction of total flavonoids from Selaginella doederleinii Hieron and its effect on the behavior of AlCl_(3)and D⁃gal⁃induced dementia mice.Methods:Whole herbs of S.doederleinii(5.5 kg)were used to extract for our study.The total flavonoids of S.doederleinii were prepared through solvent reflux,macro⁃porous resin column chromatography and other modern extraction and separation technology.Eight⁃week old Kunming male mice were divided into control group(control),dementia model group(D⁃gal,120 mg/kg,i.p.+AlCl_(3),10 mg/kg,i.g.,D⁃gal+Al)and treatment group(SLJB⁃HT,80 mg/kg,i.g.+D⁃gal+AlCl_(3)).Mice in model group and treatment group were intraperitoneally injected with D⁃galactose(120 mg/kg)and fed with AlCl_(3)(10 mg/kg)by gavage,while normal control group was intraperitoneally injected with normal saline(NS)and oral distilled water,once a day.From the 4th week,mice in the treatment group were fed with total flavonoids suspension of S.doederleinii(80 mg/kg)by oral gavage every day,and the other two groups were given distilled water by gavage once a day for 4 weeks.Anxiety⁃like behavior,recognition memory and spatial memory of animals were examined by open⁃field test(OFT),the novel object recognition test(NOR)and Y maze experiment,respectively after the last administration.Results:(1)The content of total flavonoids in SLJB⁃HT was 43.3%(433 mg/g).(2)Combined administration of D⁃gal and AlCl_(3)(D⁃gal+Al)can induce a mouse model of dementia.Compared to the control group,the model group mice showed reduced activity in the central zone of the open field.However,the SLJB⁃HT group mice exhibited significantly more activity in the central zone compared to the dementia mice.Additionally,the treatment group mice had a significantly higher percentage of distance traveled in the central zone compared to the model group mice(P<0.05),indicating a lower level of anxiety in the treatment group mice.(3)Compared to the control group and the treatment group,the model group m
