急性心肌梗死对心脏发育基因SOX9及血清炎性因子的影响

摘  要:目的探讨急性心肌缺血后SOX9的表达变化及与急性心肌缺血损害之间的关系。方法成年雄性大鼠随机分为两组:通过是否将冠状动脉左前降支处结扎分为假手术组及心肌梗死组,两组分别于0.5、8、24 h末检测心梗面积、线粒体超微结构、炎性指标与心梗标志物以及SOX9的表达情况。结果AMI后24 h内:与假手术组相比,AMI后心梗面积及线粒体均出现明显损伤,且随时间呈逐渐递增趋势(P<0.05);血清中hs-cTn、CK-MB、Mb及IL-6、TNF-α亦呈递增趋势(P<0.05);缺血心肌SOX9表达量下降,且与心肌损伤标志物及炎性因子呈负相关(P<0.05)。结论SOX9水平的下降可能与心肌的受损程度有关;心肌损伤标志物及炎性因子的变化可间接反映心肌的SOX9蛋白下降情况。Objective To investigate the changes of SOX9 expression after acute myocardial ischemia and the relationship between SOX9 expression and acute myocardial ischemic damage.Methods Adult male rats were randomly divided into sham operation and myocardial infarction groups.Myocardial infarction was produced by the ligation of the left anterior descending coronary artery.The cardiac infarct area,mitochondrial ultrastructure,inflammatory indexes and infarct markers and SOX9 were detected at the end of 0.5,8 and 24 h after modeling,respectively.Results Within 24 h after acute myocardial infarction(AMI),compared with the sham-operated group,the infarct area and mitochondria were significantly damaged after AMI,and the severity was gradually increasing(P<0.05);high-sensitivity cardiac troponin(hs-cTn),creatine kinase isoenzyme-MB(CK-MB),myoglobin(Mb),IL-6 and TNF-α in serum were increased(P<0.05).The expression of SOX9 in ischemic myocardium decreased and was negatively correlated with myocardial injury markers and inflammatory factors(P<0.05).Conclusion The decrease in SOX9 levels may be related to the degree of damage to the heart muscle.The changes of myocardial injury markers and inflammatory factors can indirectly reflect the damage and decrease of myocardial SOX9 protein.
