摘  要:为了初步探究冷冻干燥对口蹄疫病毒样颗粒(FMDV VLPs)稳定性及免疫原性的影响,采用冷冻干燥技术将FMDV VLPs疫苗抗原进行冻干处理,冷冻干燥的FMDV VLPs在保存3、6、12个月后分别免疫小鼠,检测免疫后小鼠的特异性抗体水平及淋巴细胞增殖水平;检测免疫小鼠的抗体亚型水平、细胞因子分泌水平、小鼠体重增长趋势、主要脏器指数及损伤情况等。结果显示,冷冻干燥前后FMDV VLPs的结构蛋白和粒径无明显改变;保存3、6、12月后免疫小鼠均能产生良好的特异性抗体及淋巴细胞增殖水平,抗体亚型及细胞因子分泌水平也有显著提高,且冻干前后无统计学差异;生物相容性分析显示,小鼠生长状况及主要脏器病理分析与PBS组相比无明显变化。结果表明,冷冻干燥FMDV VLPs在保存12个月后仍具有良好的免疫原性与生物相容性。To investigate the effects of freeze-drying on the stability and immunogenicity of footand-mouth disease virus like particles(FMDV VLPs).The FMDV VLPs vaccine antigen was freeze-dried using freeze-drying technology,and the mice were immunized with freeze-dried FMDV VLPs,which were stored for 3,6 and 12 months,the specific antibody and lymphocyte proliferation of the immunized mice were detected,and antibody subtypes,cytokine secretion,body weight,the index and injury of major organ were also measured.The results showed that the characteristics of FMDV VLPs did not change significantly before and after freeze-drying.After storage for 3,6 and 12 months,the immunized mice could produce high level of specific antibody and lymphocyte proliferation,and the secretion of antibody subtypes and cytokine were also significantly improved,and there was no statistical difference before and after lyophilization.Biocompatibility analysis results showed that there was no significant change in the growth status of mice and no pathological change in major organs compared with control group.This study confirmed that freeze-dried FMDV VLPs still have high immunogenicity and biocompatibility after 12 months of storage.
