摘  要:为探索益母草抗脑缺血再灌注损伤神经炎症反应的作用机制。该研究通过提取益母草中的总生物碱,将KM雄性小鼠随机分为假手术组(Sham)、模型组(CIRI)、益母草低剂量组(LHA-L,30mg·kg^(-1))、益母草中剂量组(LHA-M,60mg·kg^(-1))、益母草高剂量组(LHA-H,120mg·kg^(-1))。小鼠分组后即灌服相应药物,连续14d,其中Sham和CIRI组给予同体积的生理盐水。第14天给药1h后,对各组小鼠复制反复缺血再灌注损伤模型,再灌注24h,取材。ELISA(en-zyme-linked immunosorbent assay)检测小鼠血清的白细胞介素-6(interleukin-6,IL-6)、IL-1β和肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)水平;HE染色检测脑组织病理变化;尼氏染色法观察脑组织尼氏体的变化;免疫荧光法检测小鼠脑组织中GFAP的表达;RT-PCR法检测脑组织IL-1βmRNA、IL-6 mRNA、TNF-αmRNA、COX-2 mRNA的表达。研究结果表明,反复缺血可致小鼠急性脑缺血再灌注模型制备成功,益母草可降低脑缺血再灌注损伤小鼠血清IL-6、IL-1β和TNF-α水平,下调脑组织中GFAP蛋白表达,改善脑组织病理变化,减低脑组织中IL-1βmRNA、IL-6 mRNA、TNF-αmRNA、COX-2 mRNA的表达。益母草可以通过抑制神经炎症反应,促进细胞修复,从而保护脑缺血再灌注损伤的炎症损伤。The study aims to explore the mechanism of Leonuri Herba against cerebral ischemia-reperfusion injury neuroin-flammation.The total alkaloids were extracted from Leonuri Herba.The KM rice were divided into sham group,model group,high-dose Leonuri Herba group(LHA-H,120mg·kg^(-1)),medium-dose Leonuri Herba group(LHA-M,60mg·kg^(-1)),and low-dose of Leonuri Herba group(LHA-H,30mg·kg^(-1)).After grouping,mice were given corresponding drugs orally for 14 consecutive days,Sham and CIRI groups were given the same volume of normal saline.After the 24h-reperfusion administration,the rice were sacrificed for sample collection.Enzyme-linked immunosorbent assay(ELISA)was employed to detect serum in-terleukin-6(IL-6),IL-1βand tumor necrosis factor-α(TNF-α)levels.HE staining to detect the pathological changes of brain tissue.Nissl staining was used to observe the changes of nissl body in brain tissue.Immunofluorescence was used to detect the expression of GFAP in mouse brain tissue.RT-PCR was performed to detected expressions of IL-1βmRNA,IL-6 mR-NA,TNF-αmRNA and COX-2 mRNA in brain tissue.The mouse model of cerebral ischemia-reperfusion injury induced by repeated ischemia was successfully established.Leonuri Herba can reduce the serum levels of IL-6,IL-1βand TNF-αin mice with cerebral ischemia-reperfusion injury,reduce the expression of GFAP protein in brain tissue,alleviate the pathological changes of brain tissue,and reduce the expression of IL-1βmRNA,IL-6 mRNA,TNF-αmRNA and COX-2 mRNA in brain tissue.These results indicated that Leonuri Herba can protect the inflammatory injury of cerebral ischemia-reperfusion in-jury by inhibiting neuroinflammatory response and promoting cell repair.
