摘  要:目的:探讨不同生长周期的仙茅根茎中标志物仙茅苷的积累规律,同时测定多年生仙茅须根中仙茅苷的含量,为仙茅质量控制和资源充分开发利用提供依据。方法采用HPLC法,以ACE Excel5 C18-AR柱(5m,4.6 mm×250 mm)为色谱柱,以以乙腈-0.1%磷酸溶液(21:79)进行等度洗脱,流速:1.0ml/min;检测波长:285nm,测定各样品仙茅苷的含量。结果经过对HPLC图谱分析比较,不同生长年限的仙茅中仙茅苷含量差别较大,以4年生最高。3年生的次之,1年生则最低。3~4年生仙茅须根中仙茅苷平均含量达到0.390%~0.425%,4年生含量略高。结论3~4年生仙茅根茎的仙茅苷含量均达到《中国药典》(2020年版)规定,以3年及以上生长年限的仙茅作为药材原料具有一定的合理性。同时,3年及以上生长年限的仙茅须根的仙茅苷含量已经远超出了相同生长年限的根茎,仙茅须根可进一步开展资源评价,以便充分开发利用资源。Objective To explore the accumulation of Curculigoside in the rhizome of Curculigo orchioides in different growth pe-riods,and to determine the content of curculigoside in the fibrous root of Curculigo orchioides,so as to provide a basis for quality control and full development and utilization of resources of citronella.Methods The chromatographic separation was performed on ACE Excel5 C18-AR(5m,4.6×250mm)column.The mobile phase consisting of acetonitrile-phosphoric acid(0.1%)solu-tion(21:79)in gradient elution.The flow rate was 1.0 mL·min-1,and the detection wavelength was 285 nm.Results Through the analysis and comparison of HPLC spectra,the content of curculigoside in different growth years was different.The growth of 4 years is the highest,the growth of 3 years is the second,and the growth of 1 year is the lowest.Conclusion The content of curcu-ligoside in the rhizome of 3-4-year-old citronellae can meet the requirements of the Chinese Pharmacopoeia(2020 Edition),and it is reasonable to use Curculigo orchioides with 3-year and above growth years as the raw material of medicinal materials.At the same time,the content of curculigoside in the fibrous roots of Curculigo orchioides with 3 years and above growth years has far exceeded that of the rhizomes with the same growth years.Further resource evaluation can be carried out for the fibrous roots of Curculigo orchioides in order to fully develop and utilize the resources.
