摘 要:设计并合成了一种适用于极酸性环境的pH荧光探针。以咪唑并[1,2-a]吡啶作为电子受体,酚噻嗪衍生物作为电子供体,通过共轭结构连接,合成pH荧光探针Feno。该探针pKa为3.68,具有对H^(+)的高选择性响应,良好的光稳定性和抗各种金属离子和生物活性物质干扰的能力,共聚焦激光扫描显微镜图像显示该探针具有出色的细胞膜渗透性。探针Feno已成功应用于监测人宫颈癌细胞(HeLa)和大肠杆菌中极酸性环境的pH波动。A pH fluorescent probe named Feno suitable for extremely acidic environments was rationally designed and synthesized by utilizing imidazo[1,2-a]pyridine as an electron acceptor and phenolic thiazide derivatives as an electron donor,which were linked through conjugated structure.The probe exhibited a pKa of 3.68 and showed high selective response to H^(+),and great photostability and resistance to various metal ions and bioactive molecules interference.Confocal laser scanning microscopy images showed that the probe had excellent cell membrane permeability.The probe Feno was successfully applied to monitor pH fluctuations in the extreme acidity enviroment in HeLa cells and Escherichia.coli cells.
