摘  要:金纳米粒子(AuNPs)具有局域表面等离子共振性质,吸收光量子后能够产生显著的散射光信号。运用暗场显微镜研究AuNPs在细胞中的迁移,拟合高斯方程对AuNPs进行定位,研究AuNPs标记分子在细胞中的靶向定位方法。研究发现,HepG2细胞摄入AuNPs后分散于胞质中,轴向间隔1 nm采集1001张图片,拟合高斯方程可以得到单个粒子的相对空间位置。因此,功能化的AuNPs在活性分子的驱动下靶向作用于目标细胞器,暗场显微镜对该细胞器进行成像、定位。此方法可进一步用于研究活性分子与细胞的作用机理及新药的研发。Au nanoparticles display the property of localized surface plasmon resonance,which can generate significant scattered light signal after absorbing light quantum.In this research,dark field microscopy was used to study the migration of AuNPs in cells,and locate AuNPs by fiting Gaussian function.Meanwhile,the targeting method of AuNPs labeling molecules in cells was studied.1001 images were taken in the axial dimension with 1 nm interval,and then the relative spatial localization of single AuNP was got by fitting Gaussian function.It was found that AuNPs were dispersed in cytoplasm after intakeby HepG2 cell.Functionalized AuNPs target organelles with the motivation of active molecule,then the organelles can be imaged and localized.The method may further be applied in the research of mechanism between cells and active molecules andnew drug development.
