摘 要:目的:研究外感风痧颗粒指纹图谱与其体外抗炎作用的相关性。方法:建立外感风痧颗粒的高效液相指纹图谱;采用脂多糖刺激小鼠巨噬细胞RAW264.7模型,检测外感风痧颗粒对一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)释放的影响,考察其抗炎活性;采用灰色关联度法和偏最小二乘判别分析法(PLS-DA)分析外感风痧颗粒指纹图谱共有峰与其抗炎药效之间的谱效关系。结果:通过外感风痧颗粒HPLC指纹图谱分析,共确定了24个共有峰。灰色关联度分析结果显示,24个共有峰对药效贡献均较大;偏最小二乘判别法分析结果显示除峰3和峰16外,其他22个共有峰对抗炎药效贡献均较大,说明外感风痧颗粒体外抗炎作用是多组分协同作用的结果。结论:建立了外感风痧颗粒的指纹图谱及其体外抗炎活性的谱-效关系,为该药的Q-Marker研究提供参考。OBJECTIVE To study the spectrum-activity relationships of anti-inflammatory activities for Waigan Fengsha Granules.METHODS HPLC chromatogram of Waigan Fengsha Granules was established.The RAW264.7 model of mouse macrophages was stimulated by lipopolysaccharide to detect the effects of Waigan Fengsha Granules on the release of nitric oxide(NO),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6),so as to investigate the anti-inflammatory activities;the grey correlation analysis and PLS-DA were used to analyze the correlation of the spectrum peaks and the pharmacological activities in different samples.RESULTS A total of 24 common peaks were identified by HPLC fingerprint analysis for Waigan Fengsha Granules.The results of grey correlation analysis showed that the contribution of 24 common peaks to anti-inflammatory activities was significant,and the results of PLS-DA showed that the contribution of the other 22 common peaks to anti-inflammatory activities was significant except Peak 3 and Peak 16 The results showed that the anti-inflammatory effects of Waigan Fengsha granules in vitro were the results of multi-component synergistic actions.CONCLUSION The spectrum-activity relationships of HPLC chromatogram and anti-inflammatory effects in vitro were established,which provide reference for the further study on Q-Marker.
