摘 要:内皮素受体A(EDNRA)是内皮素受体家族成员。为探索EDNRA基因与猪脂肪代谢性状的相关性,本研究选取饲养日龄为180 d的健康去势藏公猪和大约克夏公猪作为试验对象,采用了荧光定量PCR(RT-qPCR)技术检测EDNRA基因在肝脏、肾脏、背脂和腹脂中mRNA相对表达量,运用Sanger测序法对EDNRA基因的5′侧翼区进行多态性分析。结果表明,EDNRA基因在2个猪种组织中的相对表达量趋势基本一致,表现为藏猪在肝脏和背脂中的相对表达量显著低于大约克夏猪(P<0.05),而在肾脏和腹脂中的相对表达量为藏猪极显著低于大约克夏猪(P<0.01),通过对单核苷酸多态性(SNP)进行筛选,筛选到4个SNP位点:g.11574354T>C、g.11574355G>A、g.11574363T>A、g.11574379T>A,且猪种中的等位基因频率存在显著差异(P<0.05)。推测EDNRA基因可能是调控脂肪代谢的关键基因,为进一步研究猪的脂肪调控机制奠定了基础。To explore the relevance of the EDNRA gene in fat metabolic traits of pigs,healthy depleted Tibetan boars and large Yorkshire boars reared at 180 days of age were selected as test subjects in this study.The 5'flanking region of the EDNRA gene was analyzed by Sanger sequencing.The results showed that the relative expression of the EDNRA gene in the tissues of the pig breeds were basically the same,with the relative expression in the liver and dorsal fat of the Tibetan pigs being significantly lower than that of the large Yorkshire pigs(P<0.05),and the relative expression in the kidney and abdominal fat of the Tibetan pigs was very significantly lower than that of the large Yorkshire pigs(P<0.O1),which indicated that the EDNRA gene might be positively correlated with fat metabolism.By screening the SNPs,four SNPs loci were identified:g.11574354T>C,g.11574355G>A,g.11574363T>A,and g.11574379T>A;and the allele frequencies in the pig breeds were significantly dferent(P<0.05).It is believed that the EDNRA gene might be a key gene for regulating fat metabolism,which lays a foundation for further research on the mechanism of regulating fat in pigs.
