摘 要:为探究环磷酸鸟苷依赖性蛋白激酶(PKG)基因在藏猪与大约克夏猪中的多态性与表达差异,选取180日龄藏猪和大约克夏猪,在DNA水平上对藏猪、大约克夏猪PKG基因3′侧翼区、5′侧翼区和蛋白质编码(CDS)区进行了单核苷酸多态性(SNP)位点筛选,同时在mRNA水平上用荧光定量PCR(RT-qPCR)方法检测PKG基因在心脏、肺脏和脾脏组织中的相对表达量。结果表明:PKG基因在3′侧翼区有一个突变位点′T157C,并在藏猪与大约克夏猪2个品种间存在极显著差异(P<0.01);在mRNA水平上PKG基因在心脏组织中的相对表达量表现为藏猪极显著高于大约克夏猪(P<0.01),肺脏组织中的相对表达量为藏猪显著高于大约克夏猪(P<0.05),但在脾脏组织中的相对表达量为大约克夏猪极显著高于藏猪(P<0.01)。研究表明,PKG基因可能与猪低氧适应性有关,为进一步探讨猪低氧适应性能奠定了基础。This study aimed to explore the polymorphism and expression differences of the cGMP dependent protein kinase(PKG)gene between Tibetan and large Yorkshire pigs.180-day-old Tibetan and large Yorkshire pigs were selected,and the single nucleotide polymor-phism(SNP)sites in the 3'flanking region,5'flanking region and CDS region of the PKG gene in the two breeds of pigs at the DNA level were screened.At the same time,the relative expression of the PKG gene in the heart,lung and spleen tissues of the animals were detected by fluorescence quantitative PCR(RT-qPCR)at the mRNA level.The results showed that there was a mutation site'T157C in the 3'flan-king region of the PKG gene,and there was a significant difference between Tibetan and large Yorkshire pigs(P<0.01)in this respect.In the heart tissue,the expression of the PKG gene in the Tibetan pigs was significantly higher than that in the large Yorkshire pigs(P<0.O1).In the lung tissue,the expression of the PKG gene in the Tibetan pigs was significantly higher than that in the large Yorkshire pigs(P<0.05).And in the spleen tissue,the expression of the PKG gene in the large Yorkshire pigs was significantly higher than that in the Tibetan pigs(P<0.01)at the mRNA level.It was speculated that the'T157C locus might be caused by the inconsistent expression of the PKG gene in the two pig breeds,and the gene might be related to the adaptability of the pigs to hypoxia,playing a positive role in improving this adaptability.The results of this study provided theoretical support for the follow-up study on the mechanism of the PKG gene in hypoxia adaptation of pigs.
