摘　　要：本研究旨在解析贵州白山羊肌肉生长抑制素(MSTN)基因的结构特性,为该基因功能探究提供理论依据。基于生物信息学及比较基因组学方法,对NCBI数据库中已登录的贵州白山羊MSTN基因完整编码区序列(CDS)核苷酸组成特性及编码蛋白的相关结构开展预测及分析,构建了15个物种MSTN基因系统进化树。结果表明,贵州白山羊MSTN基因CDS长度为1128bp,共编码375个氨基酸,由3个外显子和2个内含子构成,其AT含量均高于GC。贵州白山羊MSTN蛋白属混合型二级结构,存在无规则卷曲、延伸链和α-螺旋,为弱碱不稳定亲水蛋白质;包括1个18个氨基酸的潜在信号肽,无跨膜区,形成5对二硫键,含有16个磷酸化位点;278~375位氨基酸为转化生长因子-β样结构域。贵州白山羊MSTN基因与绵羊、牛等牛科动物的亲缘关系较近,与鸡形目原鸡和绿头野鸭的亲缘关系较远。本研究可为贵州白山羊MSTN基因分子遗传学及其表达调控研究奠定理论基础。In order to analyze the structural characteristics of myostatin(MSTN)gene in Guizhou white goat and provide theoretical basis for exploring its function.The nucleotide composition of the complete coding region sequence and the related structure of the encoded protein of MSTN gene in Guizhou white goat registered in NCBI database were predicted and analyzed.The phylogenetic tree of MSTN gene among 15 species was constructed based on bioinformatics and comparative genomic methods The results showed that the length of coding region of MSTN gene in Guizhou white goat was 1128 bp,encoding 375 amino acids,including three exons and two introns,and the AT content was significantly higher than that of GC.Further analysis revealed that the mixed secondary structure of MSTN protein was mainly composed of random coil,extend strand and a-helix,which was a weakly alkaline unstable hydrophilic protein.The MSTN protein of Guizhou white goat contained one signal peptide composed of 18 amino acids,no transmembrane region,forming 5 disulfide bonds,including 16 phosphorylation sites.278~375 amino acids are transforming growth factor-β-like domains.The MSTN gene of Guizhou white goat is closely related to sheep and cattle,and is relatively distant to chicken and mallard.The research would lay the foundation for molecular genetics and expression study of MSTN gene in Guizhou white goat.
