HPLC法测定柴胡舒肝丸(大蜜丸)中香附烯酮和α-香附酮的含量

摘　　要：本研究建立了柴胡舒肝丸中香附烯酮和α-香附酮含量测定方法。利用ZORBAX SB-C18(4.6 mm×250mm,5μm)色谱柱,以甲醇-水(64:36)作为流动相,流速为1.0mL/min,检测波长为254nm,柱温30℃,进样体积5μL。采用HPLC法,测定香附烯酮和a-香附酮含量分别在0.211~2.112μg(R^(2)=1.000)和0.080~0.804μg(R^(2)=1.000)的范围呈良好线性;香附烯酮和α-香附酮的精密度、稳定性(24 h)、重复性试验的RSD均小于2.0%(n=6);加样回收率分别为94.9%(RSD=2.0%)和98.1%(RSD=1.6%)。测定样品4批,香附烯酮的含量为0.10~0.41 mg/丸,α-香附酮含量为0.19~0.28mg/丸。本方法简单易操作,专属性和准确度良好,为提高柴胡舒肝丸的质量标准提供参考依据。This study established a method for the cyperotundone and a-cyperone in Chaihushugan big honeyed pills,we vtilized a ZORBAX SB-C18(4.6 mm×250 mm,5μm)column of with mobile phase consisting of methanol and water(64:36),the detection wavelength was 254 nm,the flow rate was 1.0 mL/min,the column temperature was 30 C and the injection volume was 5μL HPLC analysis was performed on.the good liner relationship of cyperotundone andα-cyperone were in the range of 0.211~2.112μg(R^(2)=1.000),0.080~0.804 pg(R^(2)=1.000),respectively.The RSD of the precision,stability(24 h)and repeatability tests of the cyperotundone and a-cyperone were all less than 2.0%.The average recoveries were 101.9%(RSD=1.7%)and 102.7%(RSD=1.6%),respectively.The content of 4 batches sample for cyperotundone and a-cyperone were 0.10-0.41 mg/pills and 0.19~0.28 mg/pills.The method is fast,accurate,reproducible,and stable,and can provide the technical support for the quality control and evaluation of Chaihushugan pills.
