时间与功率对BL21大肠杆菌超声破碎效果的影响

摘  要:以BL21大肠杆菌为实验宿主菌,研究时间与功率对BL21大肠杆菌超声破碎效果的影响,设置不同时间和功率梯度对大肠杆菌BL21进行超声破碎,并获取其胞内蛋白质,通过蛋白质浓度检测及考马斯亮蓝(Bradford法)测定蛋白质含量,进而评价大肠杆菌BL21的超声破碎效果。研究结果表明,菌液体积100 mL,菌液浓度为3.772×10^(9) cfu/mL(OD600_(nm)=1.886)时,超声功率对大肠杆菌BL21破碎效果有明显影响;破碎功率固定为5%时,破碎效果与破碎时间呈正比;破碎时间固定为1 min时,破碎功率在1%、3%和9%时其超声破碎效果逐渐提升,证明破碎效果与破碎功率呈线性关系。In this study,BL21 E.coli was used as the experimental host strain to study the effect of time and power on its ultrasonic fragmentation.BL21 E.coli was ultrasonically disrupted at different time and power gradients,and its intracellular proteins were obtained.The protein content was measured by protein concentration detection and Bradford method,and then the ultrasonic fragmentation effect of BL21 E.coli was evaluated.The results showed that the volume of bacterial solution was 100 ml and the concentration of bacterial solution was 3.772×10^(9) cfu/ml(OD600_(nm)=1.886),the ultrasonic power had a significant effect on the crushing effect of BL21 E.coli.When the crushing power is fixed at 5%,the crushing effect is proportional to the crushing time.When the crushing time is fixed at 1 min,the ultrasonic crushing effect is gradually improved when the crushing power is 1%,3%and 9%,which proves that the crushing effect is linearly related to the crushing power.The results of this study provide reference value for the research and application of ultrasonic disruption of E.coli.
