摘 要:目的探究川崎病(Kawasaki disease,KD)患儿信号转导与转录活化因子3(signal transducer and activator of transcription 3,STAT3)-细胞因子信号转导抑制因子3(suppressors of cytokine signaling 3,SOCS3)信号通路的表达及其对冠状动脉病变(coronary artery lesion,CAL)的影响。方法选取2021年1月~2022年6月住院的KD患儿50例为研究对象,根据心脏彩超结果分为CAL组(n=12)与无CAL组(n=38),另选取同期进行常规体检的健康儿童30例为对照组。分别采用实时荧光定量聚合酶链反应(real-time quantitative polymerase chain reaction,RT-qPCR)和Western blot法检测外周血单核细胞(peripheral blood monouclear cells,PBMC)中STAT3、SOCS3、基质金属蛋白酶9(matrix metalloproteinase 9,MMP9)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的mRNA和蛋白表达,酶联免疫吸附试验检测血浆中MMP9、VEGF水平。结果KD组急性期PBMC中STAT3、SOCS3、MMP9、VEGF的mRNA和蛋白表达及血浆MMP9、VEGF水平均高于KD组缓解期和对照组(P<0.05),KD组缓解期这些指标也明显高于对照组(P<0.05)。与无CAL组患儿比较,CAL组患儿PBMC中STAT3、SOCS3、MMP9、VEGF的mRNA和蛋白表达及血浆MMP9、VEGF水平均明显升高(P<0.05)。Pearson相关性分析结果显示,KD组急性期PBMC中STAT3、SOCS3的mRNA和蛋白表达分别与MMP9、VEGF呈正相关(P<0.05)。结论KD患儿STAT3-SOCS3信号通路的被异常激活,可能通过上调MMP9、VEGF表达导致CAL。Objective To investigate the expressions of signal transducer and activator of transcription 3(STAT3)-suppressors of cytokine signaling 3(SOCS3)signaling pathway in Kawasaki disease(KD)children and its effect on coronary artery lesion(CAL).Methods Fifty KD children hospitalized from January 2021 to June 2022 were selected as the study objects,and were divided into the CAL group(n=12)and the non-CAL group(n=38)according to the results of the echocardiogram examination.At the same time,30healthy children were selected as the control group.The mRNA and protein expressions of STAT3,SOCS3,matrix metalloproteinase 9(MMP9)and vascular endothelial growth factor(VEGF)in peripheral blood monouclear cells(PBMC)were examined by real-time quantitative polymerase chain reaction(RT-qPCR)and Western blot.Enzyme-linked immunosorbent assay was used to detected the plasma level of MMP9,VEGF in KD children and healthy children.Results The mRNA and protein expressions of STAT3,SOCS3,MMP9,VEGF in PBMC and the plasma levels of MMP9,VEGF in KD children in acute stage were higher than those in KD children in remission stage and control group(P<0.05).The level of all the items in KD children in remission stage were higher than those in control group(P<0.05).Compared with non-CAL group,the mRNA and protein expressions level of STAT3,SOCS3,MMP9,VEGF in PBMC and the plasma levels of MMP9,VEGF in CAL group were significantly higher(P<0.05).The results of Pearson correlation analysis showed that the mRNA and protein expressions level of STAT3 and SOCS3 in PBM of CKD children were positively correlated with MMP9 and VEGF,respectively(P<0.05).Conclusion The abnormal activation of STAT3-SOCS3signaling pathway in KD children may cause CAL by upregulating the expression of MMP9 and VEGF.
