摘 要:目的:探究低氧预适应(HPC)对氧糖剥夺/复糖复氧(OGD/R)小鼠海马神经元细胞的保护作用及其作用的相关基因。方法:将小鼠海马神经元细胞系HT22细胞分为常氧组、OGD/R组和HPC+OGD/R组。采用MTS法检测细胞活力,RT-qPCR及蛋白免疫印迹技术检测N-甲基-D-天冬氨酸受体2B亚基(NR2B)及突触后密度蛋白95(PSD95)基因的表达。结果:与常氧组比较,OGD/R组的细胞活力明显降低。HPC+OGD/R组细胞活力高于OGD/R组,细胞NR2B的表达低于OGD/R组,细胞PSD95的表达高于OGD/R组。结论:HPC对OGD/R的HT22细胞具有保护作用,其作用可能与降低NR2B的表达,升高PSD95的表达有关。Objective:To investigate the protective effects of hypoxia preconditioning(HPC)on oxygenglucose deprivation/reoxygenation(OGD/R)-induced injury mouse hippocampal neuron cells(HT22 cells)and its relevant action genes.Methods:Mouse hippocampal neuron cell line HT22 cells were divided into 3 groups:normoxic group,OGD/R group and HPC+OGD/R group.The cell viability of each group was determined by MTS method,and the expressions of N-methyl-D-aspartic acid receptor 2B subunit(NR2B)and postsynaptic density protein 95(PSD95)were determined by RT-qPCR and Western blot.Results:Compared with the normoxic group,the cell viability of OGD/R group was significantly reduced.The cell viability of the HPC+OGD/R group was significantly higher than that of the OGD/R group,and the expression of NR2B in HPC+OGD/R group was significantly lower than that in OGD/R group,and the expression of PSD95 in HPC+OGD/R group was significantly higher than that in OGD/R group.Conclusion:HPC may protect OGD/R-induced injury HT22 cells.The mechanism may be related to the reduced NR2B expression level,and the improved PSD95 expression.
