摘 要:针对某集约化养猪场,采用实时荧光定量PCR技术探究了3种典型病毒,包括猪圆环病毒2型(PCV2)、猪轮状病毒(PORV)和猪细小病毒(PPV),在污水、粪便及周边环境中的分布特征,并解析了污水处理设施对病毒的削减规律.结果显示,3种病毒在猪舍排水和粪便中普遍检出,其中PCV2浓度最高(猪舍排水中为3.31×10^(5)~7.08×10^(7)copies·mL^(-1)、粪便中为1.70×10^(3)~2.51×106copies·g^(-1)),PPV和PORV浓度较低;污水处理工艺对目标病毒的削减率为1.29~5.70 log10;养殖场周边湿地和施肥土壤中目标病毒均有检出,且浓度与猪舍排水、粪便相当,说明病毒可通过污水排放或施肥等途径向环境中传播并累积.This study applied real-time fluorescent quantitative PCR to investigate the distribution of three typical viruses,including Porcine Circovirus 2(PCV2),Porcine Rotavirus(PORV),and Porcine Parvovirus(PPV),in the wastewater,manure and surrounding environment of an intensive pig farm.In addition,the removal efficiencies of target viruses by the wastewater treatment facilities were analyzed.The results show that all three viruses were detected in swine wastewater and manure.PCV2 presented the highest abundance(3.31×10^(5)~7.08×10^(7) copies·mL^(-1) in raw wastewater and 1.70×10^(3)~2.51×10^(6) copies·g^(-1) in manure),when compared with that of PPV and PORV.The virus removal rates by the wastewater treatment process were 1.29~5.70 log10.Viruses were also detected in wetland and fertilized soil samples around the farm,with abundance comparable to those in raw wastewater and manure.This result indicates that the viruses originated from piggeries may spread and accumulate in the environment through sewage discharge or fertilization.
