摘 要:目的探讨延胡索乙素(THP)对大鼠下肢缺血再灌注(I/R)损伤的影响及作用机制。方法24只SD大鼠按照随机数字表法分为对照组、I/R组、THP组,每组8只。THP组每天分别按THP 60 mg/kg剂量连续灌胃1周,对照组、I/R组予等量0.9%氯化钠溶液连续灌胃1周。除对照组外,其余2组均构建大鼠下肢I/R模型。I/R 24 h后处死大鼠,采集血清和下肢骨骼肌组织样本;观察并比较3组大鼠下肢骨骼肌组织病理学改变;比较血清肿瘤坏死因子-α(TNF-α)、白细胞介素1β(IL-1β)、肌酸激酶(CK)水平,以及骨骼肌组织丙二醛(MDA)含量和TNF-α、IL-1β蛋白表达水平。结果与I/R组比较,THP组大鼠下肢I/R 24 h后骨骼肌组织肿胀明显减轻,炎症细胞浸润减少。I/R组大鼠下肢I/R 24 h后骨骼肌组织MDA含量和TNF-α、IL-1β蛋白表达水平以及血清TNF-α、IL-1β、CK水平均明显高于对照组(均P<0.05),THP组均明显低于I/R组(均P<0.05)。结论THP通过抑制炎症反应减轻大鼠下肢I/R损伤,具有抗大鼠下肢I/R损伤的保护作用。Objective To investigate the effects of tetrahydropalmatine(THP)on limb ischemia/reperfusion(I/R)injury in rats.Methods Twenty-four SD rats were randomized into three groups using random number table method:control group,I/R group,and THP group.Each group had eight rats.The rats received continuous gavage with different doses of 60 mg/kg THP or equal amounts of saline for one week.Rats limb I/R model were constructed in the other two groups except the control group.Then rats were sacrificed after 24 h of I/R.Serum samples and skeletal muscles tissues were collected.The pathology of rats limb skeletal muscle in three groups were observed and compared.Additionally,serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and creatine kinase(CK),and malonaldehyde(MDA)in skeletal muscle tissues were measured respectively.The protein expressions of TNF-αand IL-1βin the rats skeletal muscle were detected.Results Compared to the I/R group,the THP group significantly attenuated the rats limb skeletal muscle edema,and inflammatory cells infiltration were decreased after 24 h of I/R.The MDA content and the protein expressions of TNF-αand IL-1βin rat limb skeletal muscle tissues,and TNF-α,IL-1βand CK in serum were significantly higher in the I/R group than those in control group after 24 h of I/R(all P<0.05).The above indexes in THP group were significantly lower than those in I/R group(all P<0.05).Conclusion THP exerts a protective effect against rat hind limb I/R injury by inhibiting inflammatory reaction,reducing tissue damage.
