摘 要:目的研究药用昆虫喙尾琵琶甲Blaps rynchopetera成虫肠道的活性真菌,为抗菌、抗肿瘤活性天然产物的开发提供新的真菌资源。方法结合稀释涂布法和选择培养法对喙尾琵琶甲成虫肠道的真菌进行分离纯化;以9株病原菌为指示菌,通过牛津杯琼脂扩散法检测肠道真菌次级代谢产物的抗菌活性;结合形态学,通过分子生物学方法进行ITS序列比对,鉴定抗菌活性显著的真菌并构建系统发育树;采用5株肿瘤细胞,对已鉴定的菌株进行抗肿瘤活性测试。结果从喙尾琵琶甲肠道中分离获得108株真菌,其中17株真菌具有不同程度的抗菌活性;选择7株抑菌活性显著(抑菌圈直径≥14 mm)的真菌进行鉴定;研究还显示3株真菌的次级代谢产物具有广谱的抗肿瘤活性。结论喙尾琵琶甲肠道来源真菌的次级代谢产物具有一定的抗菌及抗肿瘤活性。Objective In order to provide new fungal resources for the development of antimicrobial and antitumor active natural products,the active fungi in the guts of medicinal insect Blaps rynchopetera adults were studied.Methods Fungi were isolated and purified from the gut of B.rynchopetera adults by using dilution coating method and selective culture method.The antimicrobial activities of secondary metabolites from the fungi were detected by Oxford cup agar diffusion assay with 9 pathogenic microorganism as indicator strain.ITS sequence alignment was performed to identify the fungi with significant activity through molecular biological method and the phylogenetic tree was constructed.Five tumor cell lines were used to test the antitumor activity of the identified strains.Results A total of 108 strains of fungi were isolated from the gut of B.rynchopetera,among them 17 strains showed different degrees of antimicrobial activity.Seven fungi with significant antibacterial activity(the diameter of the inhibition zone was greater than 14 mm)were selected for identification.The study also showed that the secondary metabolites of three strains of fungi had broad-spectrum antitumor activity.Conclusion The secondary metabolites of fungi from the gut of B.rynchopetera have certain antibacterial and antitumor activity.
