摘 要:为了解2022年浙江省猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)流行毒株的基因型及变异情况,采用RT-PCR方法,对浙江省不同地区收集的37份PRRSV阳性病料组织进行ORF5基因扩增,应用DNAstar和Mega7软件进行同源性和遗传变异分析。结果显示:37份阳性样品的ORF5基因核苷酸和编码氨基酸之间的同源性分别为82.1%~100%和81.1%~100%;阳性样品的ORF5基因与高致病性PRRSV毒株JXA1、经典PRRSV毒株VR2332、国内早期PRRSV毒株CH1-a、NADC30类毒株、NADC34类毒株和新型PRRSV毒株QYYZ的核苷酸同源性分别为84.1%~98.7%、83.9%~99.7%、85.2%~95.0%、85.3%~94.0%、86.4%~96.2%和82.8%~85.1%,编码氨基酸同源性分别为83.6%~99.5%、82.6%~99.0%、84.1%~93.5%、83.1%~94.5%、85.6%~95.5%和81.1%~86.6%。37份阳性样品的感染毒株均为美洲型,分属于1、5、8等3个谱系,其中谱系1(NADC30类和NADC34类)占比最高(23/37),有6份阳性样品的感染毒株具有强毒株特性。与参考毒株相比,37份阳性样品的GP5蛋白氨基酸变异主要集中于信号肽、2个高变区、B细胞抗原位点和2个T细胞抗原位点附近;与疫苗株相比,中和抗原表位存在氨基酸突变。结果表明:浙江省存在多个谱系美洲型PRRSV毒株流行,其中谱系1为优势流行毒株,部分毒株具有高致病性;不同毒株间存在一定的基因变异,与疫苗株相比流行毒株也出现了变异。因此,需要继续加强PRRSV监测,及时掌握PRRSV流行毒株的遗传变异状况,以便为PRRS防控提供参考。In order to investigate the genotype and variation of porcine reproductive and respiratory syndrome virus(PRRSV)prevalent in Zhejiang Province in 2022,37 PRRSV-positive samples collected from different regions of the province were used for ORF5 gene amplification by RT-PCR,and the homology and genetic variation of ORF5 gene sequences were analyzed by DNAstar and Mega 7 software.The results showed that the homologies of nucleotides and coding amino acids of 0RF5 gene sequences were 82.1%-100%and 81.1%-100%,respectively;the nucleotide homologies of PRRSV ORF5 genes of positive samples were 84.1%-98.7%,83.9%-99.7%,85.2%-95.0%,85.3%-94.0%,86.4%-96.2%and 82.8%-85.1%to highly pathogenic strain JXA1,classical strain VR2332,domestic strain CH1-a at early stage,NADC30-like strain,NADC34-like strain and novel strain QYYZ,respectively,and the encoding amino acid homologies were 83.6%-99.5%,82.6%-99.0%,84.1%-93.5%,83.1%-94.5%,85.6%-95.5%and 81.1%-86.6%,respectively.All the strains belonged to American serotype,falling into 1,5 and 8 lineages,respectively,with the highest proportion(23/37)in lineage 1(NADC30-like and NADC34-like strain),and the strains in 6 positive samples were with strong virulence.Compared to the reference strain,the amino acid variations of GP5 protein in 37 positive samples were mainly concentrated near signal peptides,2 hypervariable regions,B-cell antigen sites and 2 T-cell antigen sites;compared to the vaccine strain,amino acid mutations were available in neutralizing antigen epitope.In conclusion,several lineages of American PRRSV strains were prevalent in Zhejiang Province,especially lineage 1 that was dominant,and some strains were highly pathogenic;certain genetic variations were available in different strains,and prevalent strains also mutated compared to vaccine strain.Therefore,PRRS should be further monitored to timely learn any genetic variations of PRRSV prevalent strains,with a view to providing a reference for PRRS prevention and control.
