摘 要:目的:建立、优化多花兰组培快繁体系和种质资源保存方案。方法:以野生多花兰蒴果为材料,研究其种子无菌萌发、原球茎增殖和出芽、芽生根和保存方案。结果:种子所需萌发时间为60 d,萌发率为55.00%;原球茎培养30 d增殖率为90.00%,单个原球茎可分化出8个芽,分化出芽率达95.00%以上;芽保存方案生根率为92.50%、芽伸长约4.35 cm、芽增殖率约为90.66%,保存时间长达6个月;最适保存培养基中加入1.00 g·L^(-1)活性炭可保证生根率不变,生根时间缩短到30 d。结论:多花兰快繁体系可在120 d内获得组培苗,且仅使用基本培养基添加1~2种植物生长调节剂和/或水解乳蛋白,大大减少繁育成本,为其种苗工厂化生产奠定基础;保存方案保存时间长达6个月,对其野生资源的保护和保存具有重要意义。Objective:In order to establish and optimize the tissue culture rapid propagation system and the conservation scheme of germplasm resources of Cymbidium floribundum.Methods:The capsule of wild C.floribundum as material in this study,the culture scheme of aseptic germination of seeds,proliferation and budding of protocorms,rooting of buds,and preservation culture scheme were studied.Results:The germination time of seeds was 60 days,and the germination rate was 55.00%.The proliferation rate of protocorms was 90%cultured for 30 days.A single protocorm could differentiate into 8 buds cultured for 30 days,and the differentiation rate was more than 95.00%.The rooting rate of the bud was 92.50%,the bud elongation was about 4.35 cm,and the bud proliferation rate was about 90.66%in the preservation scheme,and the preservation time was up to 6 months.The activated carbon of 1.00 g·L^(-1) was added to the optimal preservation medium,which the rooting rate remains unchanged could be ensured,and the rooting time was shortened to 30 days.Conclusion:The rapid propagation system of C.floribundum could obtain tissue culture seedlings within 120 days and only use the basic culture medium to add 1 to 2 kinds of plant growth regulator and/or LH in this study,which greatly reduced the breeding cost.It laid a foun‐dation for the industrialized production of seedlings.The conservation plan was great significance to the protection and preservation of wild resources of C.multiflora.
