摘 要:肿瘤活体模型可以高度模拟人体内的肿瘤生长情况,从而评价药物在体内经过吸收、分布和代谢后的活性,因此,建立合适的临床前抗肿瘤药物研究模型至关重要。基于此,建立了一种新型鸡胚尿囊膜(chicken embryo allantoic membrane,CAM)种植肿瘤模型并应用于评价抗肿瘤药物的疗效,为开发新的抗肿瘤药物及寻找肿瘤治疗新策略提供模型基础。通过监测鸡胚孵育期间对移植瘤的排斥反应,根据成瘤率和成瘤大小,探究最佳气室转移时间、种瘤时间及不同肿瘤细胞系种瘤浓度,优化种瘤方式,以此构建鸡胚种植肿瘤模型。结果表明,鸡胚孵育第7天转移气室,孵育第8天硅胶环接种2×10^(6)个/颗HepG2或Hela细胞、1.5×10^(6)个/颗A498细胞、4×10^(6)个/颗MDA-MB-231细胞,能够建立稳定的鸡胚种植肿瘤模型。模型应用于抗肿瘤药物阿贝西利的药效评价,结果显示阿贝西利给药72 h后,给药组肿瘤随着给药浓度增加而逐渐减小,呈明显线性关系。鸡胚尿囊膜肿瘤模型或许是一种理想的抗肿瘤药物筛选模型,在药物疗效的临床前初步评价方面具有一定的应用前景。The in vivo tumor model can highly simulate the tumor growth in the human body,so as to evaluate the activity of drugs after absorption,distribution and metabolism in vivo,so it is very important to establish a suitable preclinical anti-tumor model.In this study,a new tumor model implanted with chick embryo allantoic membrane was established and applied to evaluate the efficacy of antineoplastic drugs.It provided a model basis for developing new antineoplastic drugs and finding new strategies for tumor treatment.By monitoring the rejection of transplanted tumor during chicken embryo incubation,and according to the tumor formation rate and tumor size,the optimal air chamber metastasis time,tumor planting time and tumor concentration of different tumor cell lines were explored,aiming to optimize the tumor planting mode and establish a stable tumor implantation model.The results showed that the stable chicken embryo implantation tumor model could be established by inoculating 2×10^(6)HepG2 cells or Hela cells,1.5×10^(6)A498 cells,4×10^(6)MDA-MB-231 cells by silicone ring on the 7th day after incubation.It was applied to the efficacy evaluation of the antineoplastic drug named Abemaciclib.The results showed that the tumor in the treatment group decreased gradually with the increase of drug concentration after 72 hours.There was an obvious linear relationship.It is concluded that the chick embryo allantoic membrane tumor model mey be an ideal model for screening antineoplastic drugs,and it has a good application prospect in the preliminary preclinical evaluation of drug efficacy.
