摘 要:目的探讨麻杏石甘汤含药肝孵育液对大鼠气道上皮细胞损伤的保护作用。方法按照麻黄-甘草三个配比(1∶4、4∶1、1∶1)制备麻杏石甘汤药液,通过外翻肠囊法、肝细胞短暂孵育分别制备含药肝孵育液A、B和C;以磷酸组胺复制气道上皮细胞损伤模型;造模后细胞分为模型组(完全培养基)、阳性对照组(200μmol·L^(-1)地塞米松)和实验A、B和C组(含药肝孵育液A、B和C),另取正常大鼠气道上皮细胞作为空白对照组。用流式细胞术检测总凋亡率,用荧光素DCFH-DA探针法测定细胞内活性氧(ROS)含量,用蛋白质印迹(Western blot)法检测上皮细胞表皮生长因子受体(EGFR)、磷酸肌醇-3-激酶(PI3K)蛋白的表达水平。结果空白对照组、模型组、阳性对照组和实验A、B、C组的细胞凋亡率分别为(21.29±0.94)%、(30.59±1.55)%、(23.16±1.76)%、(18.87±5.94)%、(17.93±5.26)%和(16.75±2.06)%,细胞内活性氧(ROS)水平分别为1.00±0.06、1.84±0.26、0.97±0.13、0.86±0.23、0.92±0.05和1.07±0.14,EGFR蛋白相对表达水平分别为0.69±0.04、1.01±0.10、0.48±0.01、0.57±0.16、0.30±0.07和0.34±0.05,PI3K蛋白相对表达水平分别为0.38±0.01、0.59±0.01、0.40±0.01、0.35±0.15、0.14±0.01和0.20±0.04;实验A、B和C组的上述指标与模型组比较,差异均有统计学意义(P<0.05,P<0.01)。结论麻杏石甘汤含药肝孵育液可能通过降低细胞活性氧水平,抑制细胞凋亡,调控EGFR/PI3K蛋白的表达,从而达到减轻气道上皮细胞损伤的作用。Objective To investigate the protective effect of liver incubation solution containing Maxingshigan decoction on airway epithelial cell injury in rats.Methods The Maxingshigan decoction was prepared according to the three ratios of ephedra and licorice(1∶4,4∶1,1∶1),and the drug-containing liver incubation solution A,B and C were prepared by the everted intestinal sac method.Airway epithelial cell injury model was replicated with histamine phosphate.After modeling,the cells were divided into model group(complete medium),positive control group(200μmol·L^(-1) dexamethasone)and experimental-A,-B and-C(drug-containing liver incubation solution A,B and C),and normal rat airway epithelial cells were taken as blank control group The total injury rate was detected by flow cytometry.The content of reactive oxygen species(ROS)in cells was determined by fluorescein DCFH-DA probe method.The expression of epidermal growth factor receptor(EGFR)and phosphoinositide-3-kinase(PI3K)protein in epithelial cells was detected by Western blot.Results The apoptosis rates of normal group,model group,positive control group and experimental-A,-B and-C group were(21.29±0.94)%,(30.59±1.55)%,(23.16±1.76)%,(18.87±5.94)%,(17.93±5.26)%,(16.75±2.06)%,respectively;the contents of reactive oxygen species(ROS)were(1.00±0.06),(1.84±0.26),(0.97±0.13)and(0.86±0.23),respectively;the relative expression levels of EGFR protein were(0.69±0.04),(1.01±0.10),(0.48±0.01),(0.57±0.16)and(0.30±0.07),respectively;the relative expression of PI3K protein was(0.38±0.01),(0.59±0.01),(0.40±0.01)and(0.35±0.15),respectively;there were significant differences in the above indexes between the experimental-A,-B and-C groups and the model group(P<0.05 or P<0.01).Conclusion Maxingshigan decoction may reduce the level of reactive oxygen species,inhibit cell apoptosis,and regulate the expression of EGFR/PI3K protein,so as to alleviate the injury of airway epithelial cells.
