摘  要:目的探究冬凌草甲素对肝内胆管癌细胞HuCCT1增殖和凋亡的影响。方法选取HuCCT1细胞随机分为对照组和低、中、高剂量实验组。对照组细胞正常培养,不做任何处理,低、中和高剂量实验组分别以3.75、7.50和15.00μmol·L-1冬凌草甲素处理24 h。用细胞计数实验(CCK-8)检测细胞增殖活性;用流式细胞术检测细胞周期和凋亡情况;用蛋白质印迹法检测增殖和凋亡蛋白水平。结果对照组和低、中、高剂量实验组G2期细胞数量百分比分别为(9.54±0.64)%、(25.39±1.29)%、(30.97±1.03)%和(35.06±4.06)%,凋亡率分别为(15.94±0.21)%、(16.96±0.88)%、(18.57±0.65)%和(30.61±0.98)%,磷酸化蛋白激酶B(p-AKT)表达水平分别为0.60±0.02、0.41±0.04、0.30±0.05和0.26±0.02,磷酸化哺乳动物雷帕霉素靶蛋白(p-mTOR)表达水平分别为0.28±0.01、0.26±0.01、0.13±0.02和0.12±0.01,磷酸化S6核糖体蛋白(p-RPS6)分别为0.32±0.02、0.28±0.01、0.17±0.01和0.10±0.01。低、中和高剂量实验组上述指标与对照组比较,差异均有统计学意义(均P<0.05)。结论冬凌草甲素具有抑制HuCCT1细胞增殖并诱导其凋亡的作用,其机制可能与冬凌草甲素调控AKT/mTOR信号通路有关。Objective To investigate the effects of Oridonin in inhibiting proliferation and inducing apoptosis in intrahepatic cholangiocarcinoma HuCCT1 cells.Methods HuCCT1 cells were randomly divided into control group,experimental-L,-M,-H groups.The control group was cultured normally without any treatment,while the experimental-L,-M,-H groups were treated with 3.75,7.50,and 15.00μmol·L-1 of oridonin for 24 h.Cell proliferation activity was detected by cell counting kit(CCK-8);cell cycle and apoptosis were detected by flow cytometry;the levels of proliferating and apoptotic proteins were detected by Western blot.Results The percentage of G2 cells in control group,experimental-L,-M,-H groups were(9.54±0.64)%,(25.39±1.29)%,(30.97±1.03)%and(35.06±4.06)%,respectively;apoptosis rates were(15.94±0.21)%,(16.96±0.88)%,(18.57±0.65)%and(30.61±0.98)%,respectively;the expression levels of phosphorylated protein kinase B(p-AKT)were 0.60±0.02,0.41±0.04,0.30±0.05 and 0.26±0.02,respectively;the expression levels of phosphorylated mammalian target of rapamycin(p-mTOR)were 0.28±0.01,0.26±0.01,0.13±0.02 and 0.12±0.01,respectively;the phosphorylated ribosomal protein S6(p-RPS6)were 0.32±0.02,0.28±0.01,0.17±0.01 and 0.10±0.01,respectively.The above indicators in the experimental-L,-M,-H groups were significantly different from those in the control group(all P<0.05).Conclusion Oridonin can inhibit the proliferation and induce apoptosis of HuCCT1 cells,and its mechanism may be related to the regulation of AKT/mTOR signaling pathway.
