摘 要:以驯化分离得到的一株耐盐好氧反硝化细菌Vibrio alginolyticus YT-3为研究对象,系统研究了其在不同C/N、盐度、温度以及pH下的脱氮性能。同时,以陶粒为载体,对Vibrio alginolyticus YT-3进行固定化,优化固定化条件,并初步确定固定化Vibrio alginolyticus YT-3对实际海水养殖废水的脱氮效果。结果表明:Vibrio alginolyticus YT-3在C/N=15~20、25~30℃、中性和偏碱性条件及较宽的盐度范围内(0~40‰)具备较强的脱氮能力。最优的固定化条件为陶粒粒径2~4 mm、固菌比为1:3(质量:体积,g:mL)、固定化时间24 h。与游离态相比,固定化后的Vibrio alginolyticus YT-3具有更宽的温度和pH适宜范围。经固定化Vibrio alginolyticus YT-3的脱氮处理,海水养殖废水中无机氮浓度下降至0.32 mg/L,总氮浓度下降至1.95 mg/L,氨氮、硝态氮、总氮的去除率达100%、85.03%、92.52%。With Vibrio alginolyticus YT-3 as the research object,a halotolerant aerobic denitrifier domesticated and isolated,its denitrification performance under different environmental conditions of C/N,salinity,temperature and initial pH were systematically studied.Meanwhile,Vibrio alginolyticus YT-3 was immobilized with the ceramsite as the carrier.The immobilization conditions were optimized,and the denitrification effect of the immobilized Vibrio alginolyticus YT-3 on the actual mariculture wastewater was preliminarily determined.The results showed that Vibrio alginolyticus YT-3 has efficient nitrogen removal ability under the conditions of C/N=15~20,25~30℃,neutral and weakly alkaline conditions,and a wide range of salinity(0~40‰).The optimal immobilization conditions are as follows:the ceramsite particle size is 2~4 mm,the solid-bacteria ratio is 1:3(mass:volume,g/mL),and the immobilization time is 24 h.Compared with the free Vibrio alginolyticus YT-3,the immobilized Vibrio alginolyticus YT-3 has a wider suitable range of temperature and pH.After the nitrogen removal treatment by the immobilized Vibrio alginolyticus YT-3,the concentrations of inorganic nitrogen and total nitrogen in the mariculture wastewater decreased to 0.32 mg/L and 1.95 mg/L respectively,and the removal ratios of NH4+-N,NO3--N and TN reached 100%,85.03%,and 92.52%.
