麻黄附子细辛汤配合电针介导TGF-β1/Smad通路调控钙通道改善窦房结纤维化

摘　　要：目的 探讨麻黄附子细辛汤配合电针对窦房结纤维化小鼠钙离子通道的影响及可能机制。方法 C57B6小鼠随机分为对照组(Control组)、模型组(Model组)、麻黄附子细辛汤配合电针治疗组(MFXE组);其中Model组、MFXE组饲养至18月龄进行实验,MFXE组灌胃麻黄附子细辛汤配合电针处理,持续21天。电生理仪监测各组小鼠心率;取心脏窦房结,分别进行HE、Masson染色,观察窦房结病理损伤及纤维化程度;Western blot检测Klotho、Trpv3、Trpv6蛋白的表达;Western blot与qRT-PCR方法检测TGF-β1/Smad通路相关TGF-β1、Smad2、Smad3和Smad7蛋白和mRNA的表达。结果 麻黄附子细辛汤配合电针治疗能够提高窦房结纤维化小鼠的心率,从而改善窦房结纤维化损伤,同时上调Klotho蛋白的表达,增加钙通道调节蛋白Trpv3、Trpv6蛋白的表达,抑制TGF-β1、Smad2、Smad3蛋白表达。结论 麻黄附子细辛汤配合电针通过调控TGF-β1/Smad通路降低窦房结纤维化损伤,促进窦房结纤维化小鼠的钙离子转运。Objective To investigate the effect of Mahuang Fuzi Xixin Decoction combined with electroacupuncture on calcium channels in mice with sinus fibrosis and its possible mechanism.Methods 18 months of C57B6 mice were randomly divided into control,model,Mahuang Fuzi Xixin Decoction combined with electroacupuncture treatment group(MFXE for 21 days).HE and Masson staining were performed respectively to observe the pathological damage and fibrosis of the sinus node.Western blot was used to detect the expressions of Klotho,Trpv3 and Trpv6 proteins.Western blot and qRTPCR method was used to detect the levels of TCF-β1,Smad2,Smad3 and Smad7 protein and mRNA.Results Mahuang Fuzi Xixin Decoction combined with electroacupuncture treatment increased the heart rate,improved the damage of sinus node fibrosis,up-regulated the expression levels of Klotho and calcium channel regulatory proteins Trpv3 and Trpv6,inhibited the expressions of TGF-β1,Smad2,Smad3.Conclusion Mahuang Fuzi Xixin Decoction combined with electroacupuncture reduces the damage of sinus node fibrosis and promotes calcium ion transport in mice with sinus node fibrosis via TGF-β1/Smad pathway.
