摘 要:近年来全球癌症的发病率和死亡率仍在持续上升,构成了极大的公共卫生挑战。肿瘤治疗效果和患者生存质量已成为科研人员亟待解决的重要课题。基于肿瘤微环境与正常生理环境p H值的差异,设计合成的p H敏感性聚合物前药可调控组织渗透和药物释放,有效避免早期突释,延长体内循环时间,从而优化药物的体内分布,降低药物的毒副作用,并实现细胞内药物的高效递送。结合实例对p H敏感性聚合物前药的制备工艺﹑主要类型及其应用现状进行了综述,并对其发展前景进行了展望,以期为聚合物前药的深入开发及临床应用提供参考借鉴。In recent years,the incidence and mortality of cancer are still rising globally,which poses a significant public health challenge.The efficacy of antitumor and the life quality of patients have become an important subject to researchers.Based on the different pH values between tumor microenvironment and normal physiological environment,the designed and synthesized pH-sensitive polymer prodrugs can regulate tissue penetration and drug release,effectively avoid sudden release at the early stage of delivery,prolong the circulation time of the drug in the body,optimize the drug distribution in biological organs,reduce the toxic and side effects,and achieve high-efficiency drug delivery in cells.In this paper,the preparation process,main types and application situation of pH-responsive polymer prodrugs were summarized with some practical examples,and the developing prospectswere forecasted,expecting to provide some reference for the further development and clinical application of polymeric prodrugs.
