摘  要:目的:观察七味白术散对糖尿病模型小鼠味觉和消化道甜味感受系统的影响,探讨其调节血糖的作用机制。方法:采用自发性糖尿病KK/Upj-Ay/J(KKAy)小鼠联合高脂高糖饮食制备糖尿病动物模型,根据血糖、体质量分层随机分为模型组(0.9%氯化钠溶液,10 m L·k g^(-1)·d^(-1)),利拉鲁肽组(利拉鲁肽注射液,200μg·kg^(-1)·d^(-1)),七味白术散组(七味白术散浸膏,14.4 g·kg^(-1)·d^(-1)),每组10只;另设10只C57BL/6J小鼠作为正常组(0.9%氯化钠溶液,10 mL·kg^(-1)·d^(-1))。连续干预8周,观察各组小鼠干预期间的摄食量、体质量、随机血糖,干预结束后采用HE染色法光镜下观察舌和十二指肠组织病理改变及舌组织味蕾横截面积,实时荧光定量PCR法检测舌和十二指肠甜味受体T1R2、T1R3和葡萄糖转运体SGLT-1 mRNA表达。结果:与正常组比较,模型组小鼠随机血糖、摄食量、体质量显著升高(P<0.01);发生舌乳头、味蕾、味孔数量下降,十二指肠黏膜机械屏障受损的病理改变,舌味蕾横截面积显著减少(P<0.01);舌T1R3 mRNA表达显著上调(P<0.05);舌和十二指肠SGLT-1 mRNA表达显著上调(P<0.01)。与模型组比较,七味白术散组小鼠摄食量(自2周起)、体质量(自4周起)持续下降(P<0.01),且效果优于利拉鲁肽组(P<0.01,P<0.05);8周时随机血糖显著降低(P<0.01);舌和十二指肠黏膜病理损伤修复,舌味蕾横截面积显著增加(P<0.01);十二指肠T1R2 mRNA表达显著上调(P<0.01),舌和十二指肠SGLT-1 mRNA表达显著下调(P<0.01)。结论:七味白术散能有效降低血糖,改善多食、肥胖症状,提高味觉敏感性,其机制可能与提高甜味感知功能,调节消化道T1R2、T1R3、SGLT-1 mRNA表达,恢复消化道甜味感受系统的糖调节能力有关。Objective:To study the effects of Qiwei Baizhu Powder on the taste system and digestive tract sweet taste receptors in diabetic model mice,and to explore its mechanism of action in regulating blood glucose.Methods:KKAy mice combined with a high-fat and high-sugar diet were used to prepare diabetes models.Stratified randomly according to blood glucose and body weight,they were divided into the diabetes group(0.9%sodium chloride solution,10 mL·kg(-1)·d(-1)),liraglutide group(liraglutide injection,200μg·kg(-1)·d(-1)),and Qiwei Baizhu Powder group(Qiwei Baizhu Powder extract,14.4 g·kg(-1)·d(-1)),with 10 mice in each group.Ten C57BL/6J mice were used as normal groups(0.9%sodium chloride solution,10 mL·kg(-1)·d(-1)).The daily intervention was carried out for 8 weeks.The food intake,body weight and random blood glucose of each group of mice during the intervention were observed.The pathological changes of tongue and duodenum and the cross-sectional area of taste buds of tongue were observed under light microscopy by HE staining,and the mRNA expression of T1R2,T1R3 and SGLT-1 in tongue and duodenal tissues was detected by real-time PCR.Results:Compared with the normal group,the random blood glucose,food intake and body weight of the diabetes group were significantly increased(P<0.01),and the number of tongue papillae,taste buds and taste holes decreased,the mechanical barrier of duodenal mucosa was damaged,and the cross-sectional area of tongue taste buds was significantly reduced(P<0.01).The expression of tongue T1R3 mRNA expression significantly increased(P<0.05),and the expression of SGLT-1 mRNA in tongue and duodenum was significantly upregulated(P<0.01).Compared with the diabetes group,the food intake(since 2 weeks)and body weight(since 4 weeks)of the Qiwei Baizhu Powder group continued to decrease(P<0.01),the effect was better than that of the liraglutide group(P<0.01,P<0.05),and significant decrease in random blood glucose at 8 weeks(P<0.01).The pathological damage of the mucosa of the tongue and duodenum
