摘  要:目的:建立超高效液相色谱法(UPLC)指纹图谱的方法,并研究蜜百合炮制过程中样品粉末颜色与22个共有峰峰面积的相关性。方法:采用UPLC法测定蜜百合的共有峰峰面积,采用CM-5分光测色仪测定所有样品粉末的色度值,对所有样品的色度值与共有峰峰面积进行Pearson相关性分析和主成分分析,确定蜜百合炮制最优时间。结果:建立蜜百合指纹图谱方法并指认王百合苷A、王百合苷B、王百合苷C、王百合苷D、王百合苷F 5个峰,其线性关系、精密度、稳定性、重复性均良好;通过色度值与峰面积相关性分析得知,蜜百合随炮制时间的延长,L^(*)值降低,a^(*)、b^(*)值升高。王百合苷A、王百合苷B与a^(*)值、b^(*)值呈正相关,王百合苷C、王百合苷D与a^(*)值、b^(*)值均呈负相关,王百合苷F与a^(*)值呈负相关,与b^(*)值均呈正相关。结论:建立蜜百合指纹图谱的UP L C方法简便可行,蜜百合炮制过程样品粉末色度值与共有峰之间具有一定的相关性,确定炮制时间最优为12 min。Objective:To establish a new method of UPLC fingerprint and study the correlation between the color of sample powder and 22 common peaks during the prepared Lilii Bulbus.Methods:UPLC was used to determine the common peak of prepared Lilii Bulbus,and CM-5 spectrophotometer was used to determine the chromaticity values of all sample powders.Pearson correlation analysis and principal component analysis were performed on the chromatic value of all samples and the common peak area to determine the optimal processing time of prepared Lilii Bulbus.Results:The fingerprint method of prepared Lilii Bulbus was established and five peaks of regaloside A,regaloside B,regaloside C,regaloside D and regaloside F were identified.The linear relationship,precision,stability and repeatability were good.Correlation analysis between chromatic value and peak area showed that L^(*)value decreased and a^(*),b^(*)value increased with processing time.Regaloside A and B were positively correlated with a^(*)and b^(*)values.Regaloside C and D were negatively correlated with a^(*)and b^(*)values.Regaloside F was negatively correlated with a^(*)and positively correlated with b^(*)values.Conclusion:The UPLC method for establishing the fingerprint of prepared Lilii Bulbus is simple and feasible.There is a certain correlation between the chromatic value of the sample powder and the common peak in the processing process of prepared Lilii Bulbus,and the optimal processing time is 12 min.
