摘  要:目的:探究蒲地蓝牙膏抗炎及促口腔黏膜溃疡愈合作用与机制,为其进一步临床应用提供理论依据。方法:将112只小鼠分为空白组、假手术组、模型组、桂林西瓜霜250 mg·kg^(-1)·d^(-1)组及蒲地蓝牙膏50、200、400 mg·kg^(-1)·d^(-1)组,每组16只。构建小鼠口腔黏膜炎模型,并检测血清中白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、前列腺素2(PGE2)等炎症因子水平;构建口腔溃疡模型,将72只小鼠分为空白组,模型组(空白赋形剂),桂林西瓜霜组(每次0.8 mg/mm^(2)),蒲地蓝牙膏低(每次0.5 mg/mm^(2))、中(每次1 mg/mm^(2))、高(每次2 mg/mm^(2))剂量组,每组12只。并以病程中溃疡状态、面积等作为评价指标,HE染色观察创面病理情况;进一步采用KB细胞炎症模型,并探究蒲地蓝牙膏抗炎、促口腔溃疡愈合的机制。结果:与模型组比较,蒲地蓝牙膏200 mg·kg^(-1)·d^(-1)、400 mg·kg^(-1)·d^(-1)组能显著降低LPS及CSE诱导的小鼠口腔黏膜炎症因子IL-6、TNF-α、PGE2水平(P<0.01,P<0.05),且蒲地蓝牙膏400 mg·kg^(-1)·d^(-1)组抗炎效果与桂林西瓜霜组相似。蒲地蓝牙膏中、高剂量组溃疡面积改善效果显著且优于桂林西瓜霜组(P<0.01),蒲地蓝牙膏各剂量组可以显著下调KB细胞IL-6 mRNA表达水平(P<0.05,P<0.01),下调炎症蛋白iNOS、COX2、NF-κB p65表达水平并能抑制IL-6、TNF-α、PGE2炎症因子的释放(P<0.01)。结论:蒲地蓝牙膏具有显著的抗炎与促伤口愈合作用,其作用机制可能是通过介导NF-κB通路从而减轻炎症反应。Objective:To explore the efficacy and mechanism of anti-inflammatory and promoting oral mucosa ulcer healing of Pudilian(PDL)tooth paste,and to provide theoretical basis for its further clinical application.Methods:A total of 112 mice were divided into blank group,sham operation group,model group,Guilin Watermelon Cream 250 mg·kg^(-1)·d^(-1)group,PDL tooth paste 50,200,400 mg·kg^(-1)·d^(-1)group,16 mice in each group.Mouse oral mucatitis model was constructed,serum levels of interleukin-6(IL-6),tumor necrosis factor-α(TNF-α),prostaglandin E2(PGE2)and other inflammatory factors were detected.The oral ulcer model was established and 72 mice were divided into blank group,model group(blank excipient),Guilin Watermelon Cream group(0.8 mg/mm^(2)each time),PDL tooth paste low(0.5 mg/mm^(2)each time),medium(1 mg/mm^(2)each time)and high(2 mg/mm^(2)each time)dose groups,12 mice in each group.The status and area of ulcers during the course of the disease were used as evaluation indexes,and the pathological conditions of the wound were observed by HE staining.Further,KB cell inflammation model was used to explore the mechanism of anti-inflammation and promoting healing of oral ulcer with PDL tooth paste.Results:Compared with the model group,PDL tooth paste 200 mg·kg^(-1)·d^(-1)and 400 mg·kg^(-1)·d^(-1)groups could significantly reduce the levels of LPS and CSE-induced oral mucosal inflammatory factors IL-6,TNF-αand PGE2(P<0.01,P<0.05).The anti-inflammatory effect of PDL tooth paste 400 mg·kg^(-1)·d^(-1)group was similar to that of Guilin Watermelon Cream group.The ulcer area of PDL tooth paste in medium and high dose groups was significantly improved and better than that in Guilin Watermelon Cream group(P<0.01).PDL tooth paste doses groups could significantly down-regulate the IL-6 mRNA expression level of KB cells(P<0.05,P<0.01).The expression levels of iNOS,COX2 and NF-κB p65 were down-regulated,and the release of IL-6,TNF-a and PGE2 inflammatory factors was inhibited(P<0.01).Conclusion:PDL tooth paste has s
