摘 要:目的对致病性耶尔森氏菌中的前噬菌体的分布情况及其基因组特征、毒力基因进行分析研究,以前噬菌体的角度,为进一步研究噬菌体与细菌的共进化关系提供线索.方法以具有完整染色体以及质粒基因组序列的82株致病性耶尔森氏菌完成图为研究对象,通过PHASTER(phage search tool enhanced release)软件对致病性耶尔森氏菌的基因组完成图进行前噬菌体预测,利用CARD(comprehensive antibiotic research database)、VFDB(virulence factors database)分析前噬菌体携带的耐药基因、毒力基因.结果82株致病性耶尔森氏菌基因组噬菌体序列携带率为100%,共预测到860个前噬菌体序列,其中完整型前噬菌体片段177个,其余683个属于噬菌体元件.每株致病性耶尔森氏菌所携带的全部前噬菌体占其基因组比例为1.7%~11.7%.35个小肠结肠炎耶尔森氏菌前噬菌体片段携带59个毒力基因,17个假结核耶尔森氏菌前噬菌体片段携带40个毒力基因,58个鼠疫耶尔森氏菌前噬菌体片段携带255个毒力基因.此外,共线性对比发现前噬菌体Haemophilus phage Aaphi23与鼠疫菌pTP33质粒高度同源且共线性好,同时发现前噬菌体Salmonella phage SSU5不但与pMT1质粒高度同源而且携带5个鼠疫菌重要毒力基因.结论82株致病性耶尔森氏菌普遍携带前噬菌体,前噬菌体是耶尔森氏菌毒力基因的重要来源,鼠疫菌前噬菌体Salmonella phage SSU5携带鼠疫菌重要毒力基因.前噬菌体Haemophilus phage Aaphi23、Salmonella phage SSU5以质粒的形式存在或是质粒的一部分.Objective Analyzing and studying the distribution,genomic characteristics,and virulence genes of prophages in pathogenic Yersinia,providing clues for further research on the co-evolutionary relationship between phages and bacteria from the perspective of prophages.Methods Taking the completed map of 82 pathogenic Yersinia strains with complete chromosomes and plasmid genomes as the research object,the genetic evolution tree of Yersinia was constructed using MUMmer,and the genome completion maps of the three pathogenic Yersinia strains were predicted using PHASTER(phase search tool enhanced release)software.CARD(comprehensive antimicrobial research database)and VFDB(virus factors database)were conducted to analyze the drug resistance and virulence genes carried by the prophage.Results The bacteriophage carrying rate of 82 pathogenic Yersinia strains was 100%,860 prophage sequences were predicted in 82 pathogenic Yersinia strains,including 177 intact prophage fragments and the remaining 683 belonging to phage originals.35 Yersinia enterocolitica prophage fragments carried 59 virulence genes,17 Yersinia pseudotuberculosis prophage fragments carried 40 virulence genes,and 58 Yersinia pestis prophage fragments carried 255 virulence genes.In addition,collinear comparison found that Haemophilus phage Aaphi23 and Y.pestis pTP33 plasmid had high homology and good collinearity.Meanwhile,it found that Salmonella phage SSU5 not only had high homology with pMT1 plasmid but also carried five important virulence genes of Y.pestis.Conclusion 82 pathogenic Yersinia strains commonly carry prophages,which is an important source of Yersinia virulence genes,and the pestis prophage Salmonella phase SSU5 carries important virulence genes of pestis.Prophage Haemophilus phase Aaphi23 and Salmonella phase SSU5 exist in the form of plasmids or are part of plasmids.
