基于超高效液相色谱-串联质谱技术对两株甘草内生菌次生代谢产物化学成分的差异分析

摘  要:参照甘草原植物对甘草中两株内生菌JTYB34和JTZB55的化学成分进行综合分析比较,对甘草内生菌次生代谢产物主要化学成分进行差异分析。采用超高效液相色谱-串联质谱(UPLC-MS/MS)技术,根据高分辨质谱提供的精确相对分子质量和碎片离子信息,结合UPLC-MS/MS检测平台和自建数据库对甘草及甘草内生菌的化学成分进行定性和定量分析。进一步结合主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)等多元统计分析方法筛选甘草内生菌JTYB34、内生菌JTZB55、甘草三者之间的差异成分。结果显示,超高效液相色谱-串联质谱(UPLC-MS/MS)技术共鉴定出1199个成分,且甘草内生菌JTYB34、内生菌JTZB55、甘草这三者之间化学成分存在显著差异。以甘草为参照物,大部分黄酮类、酚酸类、脂质类及有机酸类等化合物在JTYB34及JTZB55中含量呈现下降趋势,且化合物种类亦随之减少。聚类分析将甘草聚为Ⅰ类,JTYB34聚为Ⅱ类,其余聚为Ⅲ类;结合PCA、OPLS-DA分析结果,筛选出异甘草素、甘草酸胺、甘草黄酮A、甘草苷、4-羟基甘草查耳酮A等29个具有显著性差异的化合物。甘草内生菌JTYB34、甘草内生菌JTZB55及甘草这三组样品之间两两比较,在化学成分的含量和种类上均存在显著差异,这一发现可为甘草内生菌的药理活性研究提供一定的数据参考。The chemical constituents of two strains endophytes JTYB34 and JTZB55 from Radix Glycyrrhizae were analyzed and compared with those of other plants in Radix Glycyrrhizae.Using ultra-high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS)technology,based on the accurate molecular mass and fragment ion information provided by high-resolution mass spectrometry,the chemical constituents of Radix Glycyrrhizae and its endophytes were identified and quantitatively analyzed by UPLC-MS/MS.Furthermore,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to screen the differential components among JTYB34,JTZB55 and Radix Glycyrrhizae.The results showed that a total of 1199 compounds were identified by high-performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).There were significant differences in the chemical constituents among the endophytes JTYB34,JTZB55 and Radix Glycyrrhizae.Using Radix Glycyrrhizae as a reference substance,the contents of most flavonoids,phenolic acids,lipids and organic acids in JTYB34 and JTZB55 were decreased,and the types of compounds were also decreased.Radix Glycyrrhizae,JTYB34 and others were clustered into ClassⅠ,ClassⅡand ClassⅢrespectively,twenty-nine compounds with significant differences were screened,including isoliquiritigenin,glycyrrhizin,glycyrrhizin A,4-hydroxyglycyrrhizin chalcone A,etc.The contents and species of chemical components in three groups of samples were significantly different among JTYB34,JTZB55 and Radix Glycyrrhizae,this finding may provide some data for the study of pharmacological activity of endophytes of Radix Glycyrrhizae.
