摘 要:应用2对微卫星引物对云南17个桑粉虱种群377个雌性个体的核基因组进行扩增,分析其遗传多样性与遗传分化状况。PopGene1.32和NTSYSpc2.10e软件分析结果表明:云南桑粉虱整体水平的Nei’s基因多样性指数为0.262 1±0.224 6,Shannon’s信息指数为0.389 1±0.301 2,表现出较高的遗传多样性。17个桑粉虱种群中红河LQ种群、保山WGJ种群具有较高的遗传多样性,而曲靖DP种群、昭通ZX种群、楚雄SB种群的遗传多样性最低。UPGMA聚类分析显示,当遗传一致度为0.964时,17个桑粉虱种群聚为9个支系。其中,红河CB种群、保山种群与普洱JD种群聚为一支;曲靖LL、PJ种群与普洱ZY种群聚为一支;曲靖DP种群与楚雄SB种群聚为一支;昭通QJ种群与楚雄DY种群聚为一支;红河LQ种群、德宏ZN种群、大理HQ种群、昭通ZX种群、楚雄YR种群分别为独立支系。种群间基因分化指数Gst为0.575 0,说明17个桑粉虱种群间遗传分化程度很高。Arlequin3.1软件Mantel test分析表明,云南桑粉虱种群遗传距离与地理距离无相关性。The genetic diversity and genetic differentiation of Pealius mori about 377 female individuals from 17 popula⁃tions in Yunnan Province were studied by amplifying their nuclear genomes with two pairs of microsatellite primers.Results analysis from the software PopGene1.32 and NTSYSpc2.10e demostrated that the Nei’s gene diversity index at the overall level of Yunnan P.mori was 0.2621±0.2246,and the Shannon’s information index was 0.3891±0.3012,which indicatied high genetic diversity.Among 17 populations of P.mori in Yunnan,the genetic diversity of Honghe LQ,and Baoshan WGJ were relatively higher,while those of Qujing DP,Zhaotong ZX and Chuxiong SB were the lowest.In UP⁃GMA cluster diagram,the P.mori populations were clustered into 9 clades when the genetic identity was 0.964.Among them,Honghe CB population,Baoshan population and Puer JD population were clustered into one unit.Qujing LL and PJ population and Puer ZY population were clustered into a single unit.Qujing DP population was clustered with Chuxiong SB population.Zhaotong QJ population was clustered with Chuxiong DY population.The population of Honghe LQ,Dehong ZN,Dali HQ,Zhaotong ZX,and Chuxiong YR were independent clades,respectively.The gene diversity index(Gst)between populations of P.mori was 0.5750,indica⁃ting a high degree of genetic differentiation between the 17 populations.Mantel test by Arlequin3.1 software indicated that there was no correlation between the genetic distance and the geographic distance of P.mori populations.
