摘  要:多肉植物红宝石(Sedeveria’Pink Ruby’)为景天科拟石莲花属植物,观赏价值高,是大众非常喜爱的品种,因受环境条件影响较大,难以满足市场需求。本研究采用植物组织培养技术,以红宝石叶片为外植体,研究其无菌叶片增殖分化与出芽、叶和芽生根培养的方案。获得无菌叶片增殖分化最适培养基为MS+6-BA(3 mg·L^(-1))+NAA(0.8 mg·L^(-1))+琼脂(7 g·L^(-1))+蔗糖(30 g·L^(-1)),增殖系数达10.75;最适生根培养基为1/2MS+IBA(2mg·L^(-1))+NAA(2 mg·L^(-1))+KT(4 mg·L^(-1))+琼脂(8 g·L^(-1))+蔗糖(30 g·L^(-1))和MS+IBA(4 mg·L^(-1))+NAA(2mg·L^(-1))+KT(0.4 mg·L^(-1))+琼脂(8 g·L^(-1))+蔗糖(30 g·L^(-1)),生根率达100%。研究结果可快速获得大量优质红宝石组培苗,减少繁育成本,为红宝石的组培快繁和新品种培育提供技术支持,为工厂化育苗提供理论基础。The Sedeveria'Pink Ruby'is a popular lotus plant with high ornamental value.But as is easy to be affected by environmental conditions,it cannot meet the market demand.In this research,the approach of proliferation,differentiation and budding of sterile leaves,and rooting of leaves and buds was studied by using plant tissue culture technique with ruby leaves as explants.The most suitable culture medium of proliferation and differentiation of sterile leaves was MS+6–BA(3 mg·L^(-1))+NAA(0.8 mg·L^(-1))+Agar(7 g·L^(-1))+Sucrose(30 g·L^(-1)).The proliferation coefficient of leaves was up to 10.75.The best rooting medium was 1/2MS+IBA(2 mg·L^(-1))+NAA(2 mg·L^(-1))+KT(4 mg·L^(-1))+Agar(8 g·L^(-1))+Sucrose(30 g·L^(-1))and MS+IBA(4 mg·L^(-1))+NAA(2 mg·L^(-1))+KT(0.4 mg·L^(-1))+Agar(8 g·L^(-1))+Sucrose(30 g·L^(-1)).The rooting rate reached 100%.The research results can quickly obtain a large number of high-quality tissue culture seedlings of Sedeveria'Pink Ruby'and reduce the breeding cost,providing technical support for the rapid propagation of tissue culture and the cultivation of new varieties,as well as a theoretical basis for industrialized seedling cultivation.
