摘 要:通过超高效液相色谱-四极杆/静电场轨道阱高分辨质谱技术(UHPLC-Q-Exactive MS)分析甜瓜蒂化学成分,推测主要色谱峰的结构.采用有机溶剂萃取法将甜瓜蒂75%乙醇提取物分为石油醚、乙酸乙酯、正丁醇和水部位.利用1,1-二苯基-2-三硝基苯肼(DPPH)、羟基(·OH)、超氧阴离子(O^(-)_(2)·)自由基清除试验、铁离子还原能力(FRAP)试验、α-葡萄糖苷酶和α-淀粉酶活性的抑制作用考察甜瓜蒂乙醇提取物及其不同极性萃取部位的体外抗氧化活性和降糖活性.经UHPLC-Q-Exactive MS检测,所得MS以及MS/MS数据结合质谱裂解规律分析,初步鉴定出35个化学成分,主要为氨基酸、有机酸和萜类化合物.甜瓜蒂提取物及其不同极性部位均具有一定的抗氧化能力,正丁醇部位抗氧化能力最强,其对DPPH、·OH、O^(-)_(2)·自由基清除能力的IC 50值分别为0.2348±0.053 mg/mL、0.1485±0.027 mg/mL、0.037±0.007 mg/mL,总还原力也明显优于其他五个组分.随着浓度增加,5种组分对α-葡萄糖苷酶和α-淀粉酶抑制率不断增加,石油醚萃取物表现出较强的体外降糖活性,其对α-葡萄糖苷酶和α-淀粉酶抑制能力的IC 50值分别为12.17 mg/mL、1.452 mg/mL.本研究系统分析了甜瓜蒂中各类成分,并进行了提取物及其萃取物成分抗氧化和降糖活性测定,其中正丁醇部位体外抗氧化活性最优,石油醚部位体外降糖活性最优.Analysis of the chemical composition of PedicellusMelo tips by ultra performance liquid chromatography-quadrupole/electrostatic field orbital trap high resolution mass spectrometry(UHPLC-Q-Exactive MS)and inference of the structure of the main peaks.The 75%ethanol extract of PedicellusMelo was separated into petroleum ether,ethyl acetate,n-butanol and water parts using organic solvent extraction.The in vitro antioxidant activity and hypoglycaemic activity of ethanolic extracts of melon tips and their different polar extracted parts were investigated using 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH),hydroxyl(·OH),superoxide anion(O^(-)_(2)·)radical scavenging assay,ferric ion reducing power(FRAP)assay,inhibition ofα-glucosidase andα-amylase activities.UPLC-Q-TOF-MS,MS and MS/MS data combined with mass spectrometric cleavage patterns led to the preliminary identification of 35 chemical components,mainly amino acids,organic acids and terpenoids.The IC 50 values for the scavenging capacity of DPPH,·OH and O^(-)_(2)·radicals were 0.2348±0.053 mg/mL,0.1485±0.027 mg/mL and 0.037±0.007 mg/mL,respectively,and the total reducing power was also significantly was significantly better than the other five fractions.The five fractions showed increasing inhibition ofα-glucosidase andα-amylase with increasing concentrations.The petroleum ether extract showed strong in vitro hypoglycaemic activity with IC 50 values of 12.17 mg/mL and 1.452 mg/mL for the inhibition ofα-glucosidase andα-amylase,respectively.In this study,the antioxidant and hypoglycemic activities of the extracts and their extracts were determined,with the best in vitro antioxidant activity in n-butanol and hypoglycemic activity in petroleum ether.
