不同浓度补阳还五汤含药血清对缺氧缺糖/复氧复糖损伤的大鼠CTX-TNA2细胞系修复作用观察

摘　　要：目的探讨不同浓度补阳还五汤含药血清对缺氧缺糖/复氧复糖(OGD/R)损伤大鼠星形胶质细胞系CTX-TNA2的修复作用。方法将处于对数生长期的CTX-TNA2细胞系分为观察组、血清组、模型组、正常组。正常组不做任何处理,常规培养;模型组弃去原培养基,加入不含糖、含钙镁的无糖培养基6 h(缺氧缺糖处理),然后把无糖培养基全部更换为完全培养基培养24 h(复氧复糖);观察组和血清组细胞缺氧缺糖处理6 h后,分别用含0.5%、1%、3%、5%、10%补阳还五汤含药血清(观察1、2、3、4、5组)或含0.5%、1%、3%、5%、10%空白血清(血清1、2、3、4、5组)的完全培养基培养24 h。采用MTS法检测细胞存活率,LDH法检测细胞乳酸脱氢酶漏出率,倒置相差显微镜观察细胞状态。结果与正常组相比,模型组细胞存活率降低,且乳酸脱氢酶漏出率升高(P均<0.01);与模型组比较,观察组细胞存活率升高(P<0.01)、乳酸脱氢酶漏出率降低(P<0.01);与血清4组比较,观察4组细胞存活率高(P<0.01)、乳酸脱氢酶漏出率低(P<0.05)。观察组细胞的胞体状态和胞间突起较模型组有所恢复,且比血清组细胞好转更显著。结论与其他浓度的补阳还五汤含药血清比较,5%补阳还五汤含药血清可修复CTX-TNA2细胞OGD/R损伤。Objective To investigate the repair effect of different concentrations of Buyang Huanwu decoction-medi⁃cated serum on rat astrocyte cell line CTX-TNA2 with oxygen-glucose deprivation/reoxygenation(OGD/R)injury.Meth⁃ods CTX-TNA2 cells were divided into the observation group,serum group,model group,and normal group.Cells of the normal group were routinely cultured without any treatment.In the model group,original medium was discarded,sug⁃ar-free medium containing calcium and magnesium was added for 6 h(oxygen-glucose deprivation treatment),then the sugar-free medium was replaced by complete medium for 24 h(reoxygenation treatment).After 6 h of oxygen-glucose de⁃privation treatment,cells of the observation group and serum group were cultured with complete medium for 24 h contain⁃ing 0.5%,1%,3%,5%and 10%Buyang Huanwu decoction-medicated serum(observation groups 1,2,3,4 and 5)or 0.5%,1%,3%,5%and 10%control serum(serum groups 1,2,3,4 and 5),respectively.Cell survival rate was detect⁃ed by MTS method,lactate dehydrogenase(LDH)leakage rate was detected by LDH method,and the status of cells was observed by inverted phase-contrast microscope.Results Compared with the normal group,the survival rate decreased and LDH leakage rate increased in cells of the model group(both P<0.01).Compared with the model group,the survival rate of cells in the observation group increased(P<0.01)and LDH leakage rate decreased(P<0.01).Compared with the serum group 4,the survival rate was higher(P<0.01)and the LDH leakage rate was lower(P<0.05)in the observation group 4.The cellular body state and intercellular processes of the cells in the observation group recovered and these im⁃provements were more significant than those in the serum group.Conclusion Compared with other concentrations of Buyang Huanwu decoction-medicated serum,5%Buyang Huanwu decoction-medicated serum may repair OGD/R injury of CTX-TNA2 cells.
