摘 要:目的:探讨橙皮苷对支原体肺炎小鼠肺部炎症因子、细胞凋亡的影响及其潜在机制。方法:将小鼠随机分为正常对照组,支原体肺炎模型组,橙皮苷低、中、高剂量(80.0、160.0、320.0 mg/kg)组和阳性对照组(阿奇霉素22.5 mg/kg)。除正常对照组外,其余各组通过鼻腔接种肺炎支原体菌株法建立支原体肺炎小鼠模型。采用干湿重法检测肺组织含水量;肺功能分析系统检测静息通气量、气道阻力和肺容积;H-E染色检测肺组织病理损伤;ELISA检测支气管肺泡灌洗液(BALF)中白细胞介素(IL)-6(IL-6)、IL-1β、肿瘤坏死因子α(TNF-α)水平;全自动血细胞分析仪检测BALF中白细胞总数及巨噬细胞、中性粒细胞和淋巴细胞数;TUNEL染色检测肺组织细胞凋亡率;免疫印迹检测Bax、Bcl-2、Toll样受体4(TLR4)、MyD88、磷酸化核因子-κB p65(p-NF-κB p65)和NF-κB p65蛋白表达水平。结果:与支原体肺炎模型组比较,橙皮苷中、高剂量组和阳性对照组肺湿/干比降低,静息通气量、气道阻力和肺容积升高,肺组织病理损伤明显改善,BALF中IL-6、TNF-α和IL-1β水平降低,BALF中白细胞总数及巨噬细胞、中性粒细胞和淋巴细胞数降低,细胞凋亡率和Bax/Bcl-2蛋白表达水平降低,TLR4、MyD88、p-NF-κB p65和NF-κB p65蛋白表达水平降低。以上差异均有统计学意义。结论:橙皮苷能改善支原体肺炎小鼠肺部炎症,抑制肺组织细胞凋亡,可能与抑制TLR4/NF-κB通路有关。Objective:To explore the effect and potential mechanism of hesperidin on pulmonary inflammatory factors and cell apoptosis in mycoplasma pneumonia mice.Methods:Ninety mice were randomly divided into six groups:normal control group,mycoplasma pneumonia model group,low-dose hesperidin group(80.0 mg/kg),medium-dose hesperidin group(160.0 mg/kg),high-dose hesperidin group(320.0 mg/kg),and positive control group(azithromycin 22.5 mg/kg).Except for the normal control group,the mice model of mycoplasma pneumonia was established by nasal inoculation of mycoplasma pneumoniae strains in other groups.After each group's treatment,the moisture content of lung tissue was measured using a wet/dry(W/D)ratio.The lung function analysis system was applied to measure resting ventilation,airway resistance,and lung volume.H-E staining was used to detect the pathological injury of lung tissue.The levels of interleukin(IL)-6,IL-1β,and tumor necrosis factor-α(TNF-α)in bronchoalveolar lavage fluid(BALF)were detected by ELISA.The numbers of white blood cells,macrophages,neutrophils and lymphocytes in BALF were detected by automatic blood cell analyzer.TUNEL staining was used to measure the apoptosis rate of lung tissue.The protein expression levels of Bax,Bcl-2,toll like receptor 4(TLR4),MyD88,p-NF-κB p65,and NF-κB p65 were detected by Western blotting.Results:Compared with the mycoplasma pneumonia model group,the W/D ratio of hesperidin medium and high dose groups and positive control group were decreased,resting ventilation,airway resistance and lung volume were increased,lung histopathological injury was significantly improved,the levels of IL-6,TNF-αand IL-1βin BALF were decreased,and the numbers of total white blood cells,macrophages,neutrophils,and lymphocytes in BALF were decreased.The apoptosis rate and the protein expression level of Bax/Bcl-2 were decreased,and the protein expression levels of TLR4,MyD88 and p-NF-κB p65/NF-κB p65 were decreased,and all the differences were statistically significant.Conclusion:Hesperidin
