无舌川西小黄菊的抗炎及促凝和抗凝活性的初步测试

摘　　要：目的:对怒族民间草药无舌川西小黄菊的总醇提取物和乙酸乙酯萃取部分进行抗炎、抗氧化及促/抗凝血活性体外活性测试,测定了其总黄酮含量,并对其精油成分进行分析。方法:以巨噬细胞RAW264.7为实验细胞株,通过检测NO浓度评估抗炎活性。通过血小板聚集实验评价诱导血小板聚集活性;通过体外凝血实验研究抗凝血活性。以紫外分光光度法测定其总黄酮含量,以DPPH法测定总黄酮的抗氧化活性。通过气相色谱-质谱联用技术分析其精油化学成分。结果:无舌川西小黄菊总醇提取物和乙酸乙酯萃取物均有显著的抗炎活性。但只显示微弱的抗凝血活性,没有显示出血小板聚集活性。其总黄酮含量较高,具有良好的自由基清除能力。从其精油中分离了37个组分,鉴定了其中30个。结论:为进一步合理开发和利用无舌川西小黄菊资源提供了数据支撑。Objective To investigate the anti-inflammatory,hemostatic,anticoagulant activities of crude extract and ethyl acetate extract of Pyrethrum tatsienense var.tanacetopsis in vitro,and to determine the content of its total flavonoids,and essential oil.Methods The inhibitory activities of different extracts on NO secretion were measured by using induced model on mouse macrophages RAW 264.7.The hemostatic activities of extracts were determined by recording their effects on platelet aggregation.The anticoagulant activities of extracts were evaluated by in vitro coagulation test.The contents of total flavonoids were determined using a colorimetric method.The antioxidant activities were evaluated by DPPH methods.The constituents of essential oil were analyzed by GC-MS.Results The both extracts showed significant anti-inflammatory and antioxidant activities,but displayed weak anticoagulant activities and no hemostatic effects were observed.The contents of total flavonoids of both extracts were high.From the essential oil,37 constitutes were isolated and 30 were identified.Conclusion Our data provide a starting point for further development of Pyrethrum tatsienense var.tanacetopsis as medical resources.
