摘  要:目的:探究MicroRNA-384在脑胶质瘤细胞株中表达变化以及临床意义。方法:选取购买于中国科学院上海细胞生物所的低分级脑胶质瘤细胞株(3例)、高分级脑胶质瘤细胞株(5例)、正常脑组织细胞株(7例)为研究样本。采用体外瞬间转染法过表达或抑制U251细胞中MicroRNA-384,应用Transwell侵袭实验测定U251细胞系表达以及U251细胞系增殖以及侵袭能力的影响,应用实时荧光定量PCR技术(Q-PCR)测定U251中MicroRNA-384的含量、不同病理分级脑胶质瘤细胞株中MicroRNA-384表达水平变化情况。结果:脑胶质瘤细胞株中MicroRNA-384的含量明显低于胶质瘤组织旁正常细胞株的含量,差异有统计学有意义(t=3.418,P<0.05)。MicroRNA-384的含量随着脑胶质瘤病理分级的升高而降低,差异有统计学有意义(t=2.847,P<0.05)。与正常脑组织细胞株比较,转染MicroRNA-384组的U251细胞增殖抑制率更高,MicroRNA-384表达受到抑制,且随着培养时间延长其促进或抑制效果更明显,差异有统计学有意义(t=3.707、4.684、5.682、7.311、7.139,P<0.05)。结论:MicroRNA-384在脑胶质瘤中呈低表达,且随着病理分级程度的升高而降低,脑胶质瘤细胞株中MicroRNA-384表达受到抑制,随之培养时间延长其促进或抑制效果更明显,且不同时间MicroRNA-384组的U251细胞增殖抑制率高于正常细胞组,其含量的测定可评估脑胶质瘤分化程度,为临床靶向治疗提供新思路。Objective:To investigate the changes in MicroRNA-384 expression in glioma cell lines and its clinical significance.Methods:Low-grade glioma cell lines(3 cases),high-grade glioma cell lines(5 cases),and normal brain tissue cell lines(7 cases)purchased from Shanghai Institute of Cell Biology,Chinese Academy of Sciences,were selected as samples for this study.MicroRNA-384 was overexpressed or inhibited in U251 cells using an in vitro instantaneous transfection method,and a Transwell invasion assay was applied to determine the effects of U251 cell line expression as well as the proliferation as well as invasion ability of the U251 cell line.Real-time fluorescence quantitative PCR(Q-PCR)was applied to determine the content of MicroRNA-384 in U251,and the changes in the expression level of MicroRNA-384 in brain glioma cell lines of different pathological grades.Results:The content of MicroRNA-384 in glioma cell lines was significantly lower than that of normal cell lines next to glioma tissues,and the difference was statistically significant(t=3.418,P<0.05).The level of MicroRNA-384 decreased with the elevated pathological grade of gliomas,and the difference was statistically significant(t=2.847,P<0.05).Compared with normal brain tissue cell lines,MicroRNA-384 expression was significantly up-regulated in U251 cells after transfection with MicroRNA-384mimics,and its promotion or inhibition effect was more obvious with the subsequent prolongation of the incubation time,and the difference was statistically significant(t=3.707,4.684,5.682,7.311,7.139,P<0.05).Conclusion:MicroRNA-384 was lowly expressed in gliomas,and decreased with the increase of pathological grading,and the expression of MicroRNA-384 was inhibited in glioma cell lines,and the promotion or inhibition effect was more obvious with the prolongation of culture time,and the inhibition rate of the proliferation of U251 cells in MicroRNA-384 group was higher than that of the normal cell group at different times,and the determination of its content could assess th
