摘 要:目的调查21个常染色体短串联重复(short tandem repeats,STR)序列基因座在云南汉族人群中的遗传多态性,为法医学个体识别及亲子鉴定提供基础数据。同时基于不同试剂盒19个重叠位点的遗传信息,探索22个中国群体遗传结构及群体间的遗传关系。方法采用AGCU EX22荧光检测试剂盒对1418份云南汉族个体的21个STR基因座及Amelogenin基因进行扩增,利用3130xl基因分析仪对扩增产物进行分型,利用GeneMapper®ID软件进行分析,统计21个STR基因座的等位基因频率和群体遗传学参数。利用Phlip3.695软件计算遗传距离,并利用SPSS 21.0、MVSP和Mega 7.0软件分别作多维尺度分析、主成分分析和系统发生分析。结果基因型分布均符合Hardy-Weinberg平衡,且各基因座之间均不存在连锁现象,21个STR基因座三联体累积非父排除概率为0.999999993,累积个体识别能力为0.99999999999999999999999993。群体遗传关系比较发现云南汉族群体和周边汉族群体有较近的遗传关系(Nei遗传距离≤0.0049),在多维尺度分析、主成分分析及进化分析中表现出极强的遗传亲和性。结论21个常染色体STR基因座的等位基因在云南汉族群体表现出高度的多态性,适合法医学个体识别和亲缘关系鉴定的实际需求。云南汉族群体与周边汉族群体拥有较近的遗传关系。Objective To investigate the genetic variations of 21 autosomal short tandem repeats(STRs)in Han population in Yunnan,to provide basic data for parent-child identification,and to explore the genetic relationship between Han and other 21 populations in Yunnan based on 19 overlapped STRs.Methods Twenty-one autosomal STRs and Amelogenin of 1418 Han individuals in Yunnan were amplified with AGCU EX22 kit.PCR products was detected using the ABI 3130XL Genetic Analyzer,and allele identification was performed via the GeneMapper®ID.Subsequently,allele frequencies and population genetic parameters for 21 STR loci were statistical analyzed.Nei’s genetic distance were calculated using Phlip3.695.Multidimensional scaling plots(MDS),principle components analysis(PCA),and phylogenetic analysis were performed using SPSS21.0,MVSP,and Mega 7.0,respectively.Results The genotype distribution conformed to Hardy-Weinberg equilibrium,and there is no linkage phenomenon between each locus.The cumulative non-paternal exclusion probability of 21 STR loci in a triad was 0.999999993,and cumulative individual discrimination ability was 0.99999999999999999999993.Comparison of population genetic relationships found a relatively close genetic relationship between the Yunnan Han population and the neighboring Han population(Nei genetic distance≤0.0049),and MDS,PCA,and phylogenetic tree demonstrated a strong genetic affinity.Conclusion The alleles of 21 autosomal STR loci exhibit high polymorphism in the Yunnan Han population,which is suitable for the practical needs of forensic individual identification and kinship identification.There is a close genetic relationship between the Yunnan Han population and the surrounding Han population.
