摘 要:对甘肃省天水市辣椒炭疽病菌进行分离、鉴定和生物学特性研究。结果表明,经致病性测定分离物LC8可以引起典型症状,为致病菌;LC8菌落呈圆形,初期乳白色绒毛状,后中央变为墨绿色,菌丝无色,有隔,有分枝,分生孢子长圆柱形,单孢,无色,内含油滴,大小(12.624~18.645)×(3.928~4.995)μm,分生孢子盘垫状,其上生无色、具分隔的分生孢子梗;经ITS、CHSI、ACT、TUB2和GAPDH基因序列分析表明,分离物LC8与斯高威尔炭疽菌(Colletotrichum scovillei)同源性为100%,在系统发育树上聚为一支,将其鉴定为斯高威尔炭疽菌(C.scovillei)。该菌菌丝最适生长温度25℃、pH 6.0和全黑暗,碳源为甘露醇,氮源为蛋白胨;最适产孢温度35℃、pH 8.0和12 h光暗交替,碳源为果糖,氮源为蛋白胨;最适孢子萌发温度30~35℃、pH 6.0和全黑暗,碳源为6-脱氧-L-甘露糖,氮源为蛋白胨。该研究结果为甘肃省天水市辣椒炭疽病的诊断及科学防治提供理论基础。Pepper anthracnose,a fungal disease affecting pepper crops,was isolated,identified in Tianshui of Gansu province to study its biological characteristics.The study found that the isolate LC8 was the pathogen,causing typical symptoms.The colony of isolated LC8 was round with miky white and fluffy at the initial stage,changing to blackish green at the center.The mycelia were colorless,with septum and branches,the conidia were long cylindrical,single spore,colorless,containing oil droplets,and the size was(12.624-18.645)×(3.928-4.995)μm,the acervulus were areatus,with colorless and separated conidiophores.The analysis of ITS,CHSI,ACT,TUB 2 and GAPDH gene sequences showed that the homology between the isolate LC8 and Colletotrichum scovillei was 100%,which clustered in the same branch on the phylogenetic tree,identifying it as C.scovillei.Further experiments were conducted to determine the optimum conditions for mycelial growth,sporulation,and spore germination.The results showed that the optimal conditions for mycelial growth were a temperature of 25℃,pH 6.0 and total darkness,with mannitol as carbon source and peptone as nitrogen source;the optimum conditions for sporulation were a temperature of 35℃,pH 8.0,12 h alternating light and dark,carbon source was fructose and nitrogen source was peptone;the optimum conditions for spore germination were temperature 30℃-35℃,pH 6.0 and full darkness,with fructose as the carbon source and peptone as the nitrogen source.Overall,these findings provide a theoretical basis for diagnosing and controlling the pepper anthracnose in Gansu.
