摘 要:为探究铃铛铁线莲杂交F_(1)代真实性及遗传多样性,提高其育种效率,本研究以6个德克萨斯组铃铛型铁线莲品种为亲本开展杂交组合试验,利用具有父本的特异性的ISSR多态性引物对其F_(1)代进行真实性鉴定和遗传多样性分析。结果表明:筛选的12条ISSR引物均可体现6组铃铛铁线莲杂交组合父本的特异性,37个杂交F_(1)代均为真杂种,F_(1)代出现父母结合带较多,部分F_(1)代出现了特异性条带,这表明F_(1)代中产生了遗传变异。单株和父母本的遗传相似系数变化范围较大,遗传多样性较为丰富;F_(1)代在基因上出现了明显的偏向某一方亲本的现象,为后续杂交亲本选择与选配提供借鉴。ISSR分子标记可有效地在铃铛铁线莲杂交后代生长早期用于鉴定其是否为真杂种,为杂交后代的选育提供分子依据。To investigate the authenticity and genetic diversity of the F_(1) generation of bell Clematis hybrids to improve the breeding efficiency,we made six hybrid combinations with different bell Clematis varieties,whose F_(1) generation was identified for authenticity and genetic diversity with ISSR primers with male parent specificity.The results showed that all 12 primers were able to show the specificity of male parents of the six hybrid combinations of bell Clematis and all the F_(1) generations were true hybrids.The F_(1) generation showed lots of di-parental bands,and some of them showed specific bands suggesting genetic variation in the F_(1) generation.The F_(1) generation and their parents displayed wide variation in genetic similarity coefficient and rich genetic diversity.The F_(1) generation showed a clear genetic bias in favour of one parent,providing a lesson for subsequent selection of hybrid parents and mapping.The ISSR markers were appliable for identification of true hybrids of the bell Clematis at an early growth stage.Our results provide a molecular foundation for the selection of hybrid progeny of the bell Clematis.
