摘  要:目的:研究蟾酥对人骨肉瘤细胞MG-63的作用效应及作用机制。方法:体外培养人骨肉瘤细胞MG-63,采用不同浓度的蟾酥含药血清(0%、2.5%、5%)进行干预,采用qPCR法检测各组c-myc、caspase-9的mRNA表达量,采用免疫荧光法检测各组整合素α4的含量并采用Western Blot法检测MEK5、Nur77的表达情况。分别采用ERK5抑制剂XMD8-92、XMD8-92联合5%蟾酥含药血清、MEK5过表达腺病毒干预人骨肉瘤细胞MG-63,采用Western Blot法检测c-myc、caspase-9及整合素α4的表达量。结果:相较于空白对照组(%),2.5%、5%蟾酥含药血清干预后的c-myc及整合素α4表达下降,caspase-9表达上升(P<0.05),且蟾酥含药血清干预后MEK5、Nur77表达下降(P<0.05),与干预浓度呈负相关;相较于5%蟾酥含药血清干预组,XMD8-92联合5%蟾酥含药血清组c-myc及整合素α4表达下降、caspase-9表达上升(P<0.05),而过表达MEK5联合5%蟾酥含药血清组的表达情况相反(P<0.05)。结论:蟾酥可以通过调控ERK5通路抑制人骨肉瘤细胞MG-63增殖、转移并促进细胞凋亡。Objective To study the effect and mechanism of MG-63 in human osteosarcoma cells.Methods In vitro culture of human osteosarcoma cells MG-63 with serum(0%,2.5%,5%),m RNA expression of c-myc and caspase-9 by qPCR,and integrin 4 by immunofluorescence and MEK5 and Nur77 by Western Blot.The ERK5 inhibitors XMD8-92,XMD8-92 and 5%toad-containing serum and MEK5 overexpression intervened in MG-63,and the expression of c-myc,caspase-9 and integrin 4 were measured by Western Blot.Results Compared with the blank control group,the expression of c-myc and integrin 4 decreased and caspase-9increased(P<0.05).The decreased expression of MEK5 and Nur77(P<0.05)after the serum serum intervention was negatively correlated with the intervention concentration.Compared with the 5%serum intervention group,XMD8-92 and caspase-9 expression of c-myc-integrin 4 increased in the 5%serum containing serum group(P<0.05),while the expression of MEK5 and 5%serum group was reversed(P<0.05).Conclusion Busu can inhibit the proliferation and metastasis of MG-63 and promote apoptosis in human osteosarcoma cells by regulating ERK5pathway.
