摘  要:目的 利用网络药理学方法探讨丹七片治疗急性心肌梗死的物质基础及作用机制。方法 检索TCMSP获得丹七片组方药物的活性成分和作用靶点。通过SwissTargetPrediction数据库检索丹七片药物靶点。利用UniProt数据库对药物靶点进行规范化处理。在DrugBank、DisGeNET数据库筛选急性心肌梗死相关靶点。通过STRING数据库进行靶点蛋白相互作用关系分析,构建蛋白质-蛋白质相互作用网络。利用DAVID数据库对疾病和药物的交集靶点进行GO功能富集分析,利用KEGG Mapper平台进行KEGG通路富集分析,运用Cytoscape3.8.0进行可视化处理和网络拓扑结构分析。结果 获得丹七片42个活性成分,核心成分包括丹参酮、木犀草素、槲皮素等,获得急性心肌梗死166个靶点,获得丹七片治疗急性心肌梗死的核心靶点23个,包括CES1、MMP9、MMP2、PTGS2等。GO功能富集结果显示,丹七片在生物过程、分子功能、细胞组分等多方面影响急性心肌梗死的发生和发展。KEGG通路富集主要涉及血管直径的调节通路、蜕膜化通路、血管内皮生长因子刺激的细胞反应等通路。结论 丹七片可能通过多成分、多靶点和多通路协同作用,发挥降低心肌内皮细胞炎性反应、减轻缺血再灌注损伤等作用,从而治疗急性心肌梗死。Objective To explore the mechanism of Danqi Tablets in the treatment of acute myocardial infarction through network pharmacology methods.Methods The TCMSP was used to acquire the main chemical components and action targets of Danqi Tablets.The drug targets of Danqi Tablets were retrieved through the SwissTargetPrediction platform.The UniProt database was used to normalize the acquired drug targets.The AMI-related targets were screened in DrugBank and DisGeNET databases.The protein interaction relationship analysis was carried out through STRING database to construct protein-protein interaction network.The DAVID platform was used to conduct GO enrichment analysis,and the KEGG Mapper platform was used to conduct KEGG pathway enrichment analysis.Visualization analysis and network topology analysis were conducted using Cytoscape 3.8.0.Results Totally 42 components of Danqi Tablets were obtained,including tanshinone,luteolin,quercetin,and 166 targets of acute myocardial infarction,and 23 core targets of Danqi Tablets for the treatment of acute myocardial infarction were obtained,including CES1,MMP9,MMP2,PTGS2,etc.The GO functional enrichment results showed that Danqi Tablets affected the occurrence and development of acute myocardial infarction in various aspects such as biological processes,molecular functions,and cellular composition.The enrichment of KEGG pathway mainly involves the regulation pathway of vascular diameter,decidualization pathway,vascular endothelial growth factor stimulated cell response and other pathways.Conclusion Danqi Tablets may play a synergistic role in reducing inflammatory response of myocardial endothelial cells and alleviating ischemia-reperfusion injury through multiple components,targets,and pathways,thus treating acute myocardial infarction.
