摘 要:[目的]本文旨在探究5℃低温处理对芹菜种子萌发的抑制作用及其调控机制。[方法]以‘上海黄心芹’种子为试验材料,设置5和20℃处理,探究其萌发情况、赤霉素(GA)和脱落酸(ABA)含量及代谢通路中关键基因表达量动态变化及淀粉及可溶性糖含量的动态变化。[结果]5℃低温处理显著抑制了芹菜种子萌发率、种子出苗率、相对吸水量和种胚的生长,20℃处理则能快速促进种子萌发,将5℃处理15 d后的种子转入20℃后(T_(5-20)处理),种子又能快速恢复萌发活力,说明5℃低温并没有对种子造成不可逆伤害,种子活力染色的结果也说明低温并没有对种胚结构造成伤害。5℃低温还显著抑制了种子萌发期间GA的合成,相关的代谢及下游基因也被明显抑制表达,而20℃处理则显著促进GA的合成,并且GA3ox1基因在20℃处理以及T_(5-20)处理中的高表达说明其可能对GA的合成发挥关键作用;5℃处理中ABA含量呈波动变化,但GA/ABA整体呈下降趋势,ABA降解基因CYP707A1的表达量与5℃处理种子萌发中后期的ABA含量有明显的负相关趋势。另外,5℃还明显抑制了芹菜种子的糖代谢能力,5℃处理的种子在萌发期的淀粉含量明显高于20℃处理,但可溶性糖含量明显低于20℃处理。[结论]5℃低温诱导芹菜种子发生了次生休眠,GA/ABA可能是芹菜种子响应环境温度的重要信号,并且GA3ox1和CYP707A1是潜在的应对低温的关键基因。[Objectives]The paper aimed to investigate the inhibitory effect of 5℃low temperature treatment on seed germination of celery and its regulatory mechanism.[Methods]The seeds of‘Shanghai yellow heart’were treated at 5 and 20℃to explore the germination,gibberellin(GA)and abscisic acid(ABA)contents,the dynamic changes of key genes expression in metabolic pathways,and the dynamic changes of starch and soluble sugar contents.[Results]5℃low temperature treatment significantly inhibited the seed germination rate,seedling emergence rate,relative water absorption and embryo growth of celery,while 20℃treatment could rapidly promote the seed germination.And after the seeds treated at 5℃for 15 days were transferred to 20℃(T_(5-20)treatment),the seeds could quickly recover their germination vitality,which indicated that 5℃low temperature did not cause irreversible damage to the seeds.The results of seed vitality staining also indicated that low temperature did not cause damage to the embryo structure.Low temperature at 5℃also markedly inhibited the synthesis of GA during germination,and the expression of related metabolism and downstream genes was also markedly inhibited,while 20℃treatment significantly promoted the synthesis of GA,and the high expression of GA3ox1 gene in seeds treated at 20℃and T_(5-20)meant that it might play a key role in the synthesis of GA.ABA content showed a dynamic change in 5℃treatment,but GA/ABA showed a downward trend in adults.The expression of the ABA degrading gene CYP707A1 had a significant negative correlation with ABA content in the middle and late stages of seed germination in 5℃treatment.In addition,5℃also significantly inhibited the sugar metabolism capacity of celery seeds.The starch content of celery seeds treated at 5℃was significantly higher than that treated at 20℃,but the soluble sugar content was significantly lower than that treated at 20℃.[Conclusions]Low temperature at 5℃induced secondary dormancy of celery seeds.GA/ABA might be an important
