摘  要:[目的]本文旨在探究亚硝酸盐对盐蚕豆虫(Fabrea salina)的胁迫机制。[方法]采用静态水生毒性试验法,研究不同质量浓度亚硝酸钠(0、9.63、19.25、38.50、77.00和154.00 mg·L^(-1))胁迫下盐蚕豆虫的种群动力学影响,并将超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)和过氧化物酶(POD)作为生物标志物,应用综合生物标志物响应指数(IBR)整合4种生物标志物。[结果]慢性毒性试验表明:随着亚硝酸钠质量浓度的增加,盐蚕豆虫最大种群密度和比生长率(μ)呈先上升后下降的趋势,在9.63 mg·L^(-1)处理组达到峰值,增加了8.8%;在154.00 mg·L^(-1)处理组达到最小值,下降了20.59%。盐蚕豆虫的世代时间则先缩短后延长。抗氧化酶活性毒性试验表明:盐蚕豆虫SOD活性随亚硝酸盐质量浓度的升高呈先上升后下降趋势;胁迫前期CAT和GPx活性随亚硝酸盐质量浓度的升高也呈先上升后下降趋势,胁迫后期GPx活性则显著升高;盐蚕豆虫POD活性随亚硝酸盐质量浓度的升高呈上升趋势。[结论]高质量浓度亚硝酸钠对盐蚕豆虫有一定的生物毒性,导致其种群生长状态变差,亚硝酸盐会导致盐蚕豆虫产生氧化应激反应。IBR指数作为量化污染物诱导的各种生物标记物反应的有效工具,可以揭示亚硝酸盐暴露对盐蚕豆虫抗氧化酶活性的影响。[Objectives]This paper aimed to explore the toxic mechanism of nitrite on Fabrea salina.[Methods]The toxicity of sodium nitrite with different concentrations(0,9.63,19.25,38.50,77.00 and 154.00 mg·L^(-1))was examined on the population dynamics of F.salina by a static bioassay test.Superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GPx)and peroxidase(POD)were used as biomarkers and integrated by the biomarker response index(IBR).[Results]The chronic toxicity experiments showed that with the increase of sodium nitrite concentration,the maximum population density and specific growth rate(μ)increased first and then decreased,and reached the peak value in the 9.63 mg·L^(-1)treatment group,increasing by 8.8%.The minimum value was reached in the 154.00 mg·L^(-1)treatment group,with a decrease of 20.59%.Generation time fell first and then rose.The toxicity test of antioxidant enzyme activity showed that,the activity of biomarkers had various trend under sodium nitrite stress,and the SOD activity of F.salina showed a trend of first increasing and then decreasing.The CAT and GPx activity in the early stage of stress also increased first and then decreased with the increase of sodium nitrite concentration,and the GPx activity increased significantly in the later stage of stress.The POD activity of F.salina showed an upward trend with the increase of sodium nitrite concentration.[Conclusions]High concentration of sodium nitrite had certain biological toxicity to F.salina,including the deterioration of the population growth state and leading to oxidation stress response.As a feasible tool to quantify pollutant-induced responses to various biomarkers,the IBR index could reveal the effects of nitrite exposure on the activities of antioxidant enzymes of F.salina.
