摘  要:铁死亡是一种较新的细胞程序性死亡形式,但不同于凋亡、自噬等其他细胞程序性死亡,其发生机制与铁过载、脂质过氧化蓄积、氨基酸代谢异常有关。全文阐述了铁死亡的触发及GPX4、辅酶Q10、FSP1、DHODH等分子调控的防御机制,总结了相关非编码RNA通过调控铁死亡来影响胃癌发生及进展的作用;在胃癌治疗方面,铁死亡途径的激活对胃癌化疗具有辅助提升效应,一些中草药被发现能介导铁死亡发挥治疗作用;总结了胃癌治疗中具有潜在价值的铁死亡靶向候选物质和基因。Ferroptosis is a newly discovered type of programmed cell death.Differing from other types of programmed cell death such as apoptosis and autophagy,its mechanism is related to iron overload,lipid peroxidation accumulation,and abnormal amino acid metabolism.In this article,the triggering of ferroptosis and defence mechanism through regulation of GPX4,coenzyme Q10,FSP1,and DHODH are described.The role of some non-coding RNAs in the occurrence and development of gastric cancer by regulating ferroptosis is summarized.The activation of the ferroptosis has an auxiliary enhancing effect on gastric cancer chemotherapy,and some Chinese herbal medicines may induce ferroptosis to play therapeutic roles.The paper also suggests some candidate targeted drugs and genes related ferroptosis as potential means for treatment of gastric cancer.
