摘  要:目的通过生物信息技术分析GSE72224基因芯片数据,比较衰老小鼠与青年小鼠B细胞基因差异,探索关键基因,为衰老机制研究及药物研发提供思路。方法通过NCBI提供的GEO数据库下载GSE72224,利用生物信息方法筛选衰老与青年小鼠B细胞差异表达基因,然后对差异表达基因进行基因本体论(GO)功能富集分析和京都基因组百科全书(KEGG)信号通路分析,并且构建蛋白质相互作用网络获得关键基因和关键模块。结果共筛选出87个表达基因,包括其中66个下调基因及21个上调基因。差异表达基因主要参与B细胞活化、内质网及内质网蛋白加工通路等。通过构建蛋白质相互作用网络从而筛选出H2A组蛋白家族成员(H2af)x、Zeste基因增强子人类同源物(Ezh)2、蛋白二硫化物异构酶(Pdi)A4、H2B组家族成员b(Hist2h2bb)、白细胞介素(IL)-10、Sel1l、Dnajb9、泛素样含PHD和环指域(Uhrf)1这8个关键基因。并且筛选出两个重要模块,模块中的基因主要也与内质网蛋白加工通路等有关。结论H2afx、Ezh2、Pdia4、Hist2h2bb、IL-10、Sel1l、Dnajb9、Uhrf1此8个关键基因,主要参与B细胞发育、DNA甲基化和内质网的蛋白质调控等过程。但是有关基因的上下游作用点还需深入研究。Objective To analyze the data of GSE72224 gene chip by bioinformatics technology,compare the gene differences of B cells of aging mice and young mice,explore the key genes,and provide ideas for research on the mechanism of aging and drug development.Methods GSE72224 was downloaded from the GEO database provided by NCBI.Bioinformatics method was used to screen the differentially expressed genes in B cells of aging and young mice.Then,gene ontology(GO)function enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)signaling pathway analysis of differentially expressed genes were performed,and protein interaction network was constructed to obtain key genes and key modules.Results A total of 87 expression genes were screened,including 66 down-regulated genes and 21 up-regulated genes.Differentially expressed genes were mainly involved in B cell activation,endoplasmic reticulum and endoplasmic reticulum protein processing pathways and so on.Eight key genes,member of the H2A histone family(H2af)x,human congener of Zeste gene enhancer(Ezh)2,protein disulfide isomerase(Pdi)A4,group H2B family member b(Hist2h2bb),interleukin(IL)-10,Sel1l,Dnajb9 and ubiquitin sample contains PHD and ring finger domain(Uhrf)1 were screened out through the construction of protein interaction network.Two important modules were screened out,and the genes in the modules were mainly related to the endoplasmic reticulum protein processing pathway.Conclusions Eight key genes,H2afx,Ezh2,Pdia4,Hist2h2bb,IL-10,Sel1l,Dnajb9 and Uhrf1 are mainly involved in B cell development,DNA methylation and protein regulation of the endoplasmic reticulum.But further research is needed on the upstream and downstream action of genes.
