摘  要:目的建立大承气汤单煎液与合煎液UPLC指纹图谱,比较大承气汤单煎液与合煎液的差异。方法采用UPLC法,以Waters ACQUITYHSS T3柱(2.1 mm×150 mm,1.8μm)为色谱柱,柱温为30℃,以甲醇-0.1%磷酸溶液为流动相,梯度洗脱,流速为0.25 mL/min,检测波长为225 nm,测定20批次大承气汤单煎液与合煎液指纹图谱,并采用相似度评价软件、主成分分析(PCA)和正交偏最小二乘判别(OPLS-DA)对结果进行分析。结果建立了大承气单煎液与合煎液UPLC指纹图谱,标定了19个共有峰,指认了其中11个峰;20批大承气汤单煎液、合煎液相似度均在0.90以上,PCA分析出4个主成分反映了19个共有色谱峰的信息;OPLS-DA分析筛选出影响大承气汤单煎液与合煎液差异的5个成分峰。结论本研究为大承气汤中药复方配方颗粒的制备工艺及质量标准的提高研究提供参考。Objective To establish the UPLC fingerprints of single decoction and mixed decoction of Dachengqi Decoction,and compare the differences between them.Methods The UPLC analysis was performed on a Waters ACQUITY®HSS T3 column(2.1mm×150 mm,1.8μm)with the column temperature set at 30℃,the mobile phase was methanol-0.1%phosphoric acid solution in gradient elution mode with a flow rate of 0.25 mL/min,and the detective wavelength was 225 nm.The fingerprints of 20 batches of single decoction and mixed decoction of Dachengqi Decoction were determined,the results of 20 batches of samples were evaluated by the similarity evaluation system,PCA and OPLS-DA.Results The UPLC fingerprints of single decoction and mixed decoction of Dachengqi Decoction was established,19 Common Peaks were calibrated,and 11 of them were identified.The similarity of UPLC fingerprints of 20 batches of single decoction and mixed decoction of Dachengqi Decoction was above 0.90,the 4 principal components in PCA analysis reflected the information of 19 common peaks,and the 5 component peaks which affected the difference between single decoction and mixed decoction of Dachengqi Decoction were screened.Conclusion The study provides a reference for the preparation technology and improvement of quality standard of compound granules of Dachengqi Decoction.
